Airway epithelial cells are the first cells to be challenged upon contact with the conidia of Aspergillus. In response, they express pattern-recognition receptors that play fundamental roles as sentinels and mediators of pulmonary innate immunity. The C-type lectin Dectin-1 is expressed predominantly on the surface of myeloid lineage cells. We examined the induction, regulation, and functions of Dectin-1 in pulmonary epithelial cells by challenging human bronchial epithelial (HBE) cells with A. fumigatus. Inflammatory, antimicrobial peptide genes and reactive oxygen species (ROS) were quantified, with and without knockdown of Dectin-1. We found that A. fumigatus induced the expression of Dectin-1 mRNA and protein in HBE cells in a toll-like receptor (TLR) 2-dependent manner. In addition, A. fumigatus-mediated generation of ROS was dependent on the upregulation of Dectin-1. Moreover, A. fumigatus actively induced the expression of TNFα, GM-CSF, IL8, HBD2, and HBD9. Knockdown of Dectin-1 inhibited TNFα, IL8, HBD2, and HBD9 expression. Hence, Dectin-1 was required for the upregulation of pro-inflammatory cytokines and antimicrobial peptides. Finally, knockdown of TLR2 significantly inhibited Dectin-1 upregulation. Our results demonstrate the novel induction of Dectin-1 in human bronchial epithelial cells and its critical role in the innate immune response against A. fumigatus in non-phagocytic cells.
Onshore and offshore currents and the associated volume transport across three isobaths (50, 100, and 200 m) over the continental shelf of the East China Sea were examined using daily reanalysis data in 1993–2012. After being averaged along the isobaths, the velocities across 100 and 50 m are onshore in the bottom layer but offshore in the surface layer. In contrast, those across the 200-m isobath are onshore in the surface and bottom layers but without a clear direction in the midlayer, suggesting a three-layer structure. The surface offshore current across the 100-m isobath mainly arises from the Taiwan Strait Current, while the surface onshore current across the 200-m isobath mainly arises from the Kuroshio, both of which converge in the area between the 100- and 200-m isobaths and flow toward the Tsushima Strait. The control of bottom Ekman dynamics on the onshore bottom currents is important at the 100-m isobath, partly important at the 200-m isobath, and slightly important at the 50-m isobath. The seasonal variations of onshore and offshore currents in the surface layers across the three isobaths are likely caused by local winds, the Taiwan Strait Current, and the Changjiang discharge, while those in midlayer across the 200-m isobath demonstrate a strong geostrophic control and can be interpreted from a traditional viewpoint on the Kuroshio intrusion over the entire water column across the shelf slope. The close connection of bottom onshore currents across the three isobaths suggests that the bottom layer is an important pathway for water exchange of shelf water and the open sea.
Uncovering the genetic basis of key agronomic traits, and particularly of drought tolerance, addresses an important priority for durum wheat improvement. Here, a genome-wide association study (GWAS) in 493 durum wheat accessions representing a worldwide collection was employed to address the genetic basis of 17 agronomically important traits and a drought wilting score. Using a linear mixed model with 4 inferred subpopulations and a kinship matrix, we identified 90 marker-trait-associations (MTAs) defined by 78 markers. These markers could be merged into 44 genomic loci by linkage disequilibrium (

> 0.2). Based on sequence alignment of the markers to the reference genome of bread wheat, we identified 14 putative candidate genes involved in enzymes, hormone-response, and transcription factors. The GWAS in durum wheat and a previous quantitative trait locus (QTL) analysis in bread wheat identified a consensus QTL locus.4B.1 conferring drought tolerance, which was further scanned for the presence of potential candidate genes. A haplotype analysis of this region revealed that two minor haplotypes were associated with both drought tolerance and reduced plant stature, thought to be the effect of linkage with the semi-dwarfing gene

. Haplotype variants in the key chromosome 4B region were informative regarding evolutionary divergence among durum, emmer and bread wheat. Over all, the data are relevant in the context of durum wheat improvement and the isolation of genes underlying variation in some important quantitative traits.
