An improved peroral intubation method is described far the study of gastric secretion in the guinea-pig. Volume, pH, titratable acidity, and output were registered under basal conditions and after stimulation with pentagastrin and betazole in doses ranging from 0.1 to 1125 pg/kg/h and 0.1 to 225 mglkg/h respectively. The method is particularly suited for long-term studies of certain pathological conditions o f the gastric mucosa.

Volume, pH, and total acidity of the basal and of the stimulated gastric secretion were measured in guinea-pigs before, and at appropriate intervals after, x-irradiation to the stomach area. A great reduction of the stimulated acidity and acid output was found. It reached a maximum between the second and the fourth week after irradiation. Thereaftcr the total acidity increased gradually. but, 56 weeks after irradiation, it still remained significantly below the normal (p < 0.05). Histological studies and parietal cell counts were carried out in guinea-pigs after x-irradiation to the stomach area. There was a pronounced reduction of the parietal cell mass, parietal cell density, and glandular mucosal volume. A reduction of a lesser degree persisted one year after irradiation, although the mucosa appeared almost normal. The histological changes were most pronounced during the period of two to four weeks after irradiation and resembled thosc seen in humans with chronic atrophic gastritis.
