Anti-stress effects of polyphenol extracts of were evaluated. Polyphenol extracts of and diazepam were compared for anti-stress activities using a cold restraint animal stress model. extracts were administered orally at dosages of 150 and 450 mg/kg. Diazepam, an anti-stress agent, was used as a standard drug at 0.18 mg/kg p.o. Both dosages of extracts showed good anti-stress effects. Due to cold restraint stress there was an imbalance in levels of biochemical parameters, including glucose, triglycerides, cholesterol, alkaline phosphatase (ALP), alanine aminotransferase (ALT), and aspartate amino transferase (AST), which were near normalized following adminstration of extracts. Polyphenol extracts of S. polycystum at oral dosages of 150 and 450 mg/kg exerted anti-stress effects.
AbstrakPenelitian ini bertujuan untuk mengetahui aktivitas antioksidan, kandungan total fenol, kandungan nutrisi, dan kandungan fitokimia dari bubuk kering Sargassum hystrix. Ekstrak air dari sampel kering alga cokelat (S. hystrix) disiapkan dan diuji kandungan total fenolnya. Jumlah kandungan total fenol ekstrak air yaitu 11,43 g Gallic Acid Equivalent (GAE)/100 g berat kering sampel dengan menggunakan metode Folin-Ciocalteu. Aktivitas antioksidan (Radical Scavenging Activity/RSA) BHT yang digunakan sebagai standar tergolong tinggi, penghambatan radikal DPPH pada 1 mg sampel kering/ml pelarut yakni 96%. Nilai IC 50 BHT adalah sebesar 0,227 ± 0,001 mg sampel kering/ml pelarut, sedangkan aktivitas penangkapan radikal (RSA) bubuk kering S. hystrix sebesar 65,28% pada 1 mg sampel kering/ml pelarut. Nilai IC 50 bubuk kering S. hystrix adalah sebesar 0,616 ± 0,005 mg sampel kering/ml pelarut. Kandungan nutrisi dan komponen bioaktif bubuk kering S. hystrix seimbang untuk mendukung aktivitas antioksidan sampel. Hasil ini menunjukkan bahwa alga tersebut potensial sebagai sumber antioksidan alami. Water extract from the dried sample of brown algae (S. hystrix) was prepared and examined for its phenolic compounds. The amount of total phenolic compounds in water extract was about 11.43 g Gallic Acid Equivalent (GAE)/100 g of dry basis sample, as measured by using Folin-Ciocalteu method. The antioxidant activity (Radical Scavenging Activity/RSA) of BHT as a standard was high about 96% inhibition of radical DPPH with 1 mg dry sample/ml solvent. The IC 50 of the BHT was 0.227 ± 0.001 mg of dry sample/ ml solvent. While the radical activity (RSA) of S. hystrix dry powder was about 65.28% at 1 mg dry sample/ml solvent. The IC 50 of S. hystrix dry powder was 0.616 ± 0.005 mg of dry sample/ml solvent. Nutrient contents and bioactive compounds of dry powder S. hystrix were balanced to support antioxidant activity of the sample. The results suggest that this alga is a potential source of natural antioxidant.
This study was conducted to determine the influence of Sargassum hystrix powder (SHP) as an alternative source of functional food for treating in vivo stress by measuring levels of glucose, triacylglycerol, total cholesterol, and cortisol, and liver histopathology. Wistar rats aged 3 months and weighing 150~200 g were divided into 7 groups: normal control, fasting control, negative control (stress without adaptogen), and 4 experimental conditions (stress+0.18 mg/kg diazepam, stress+450 mg/kg pellet, stress+mixture of pellet with SHP 450 mg/kg, and stress+450 mg/kg of SHP). Intake of liquids and and body weight were measured daily. Blood samples were collected on day 0 (baseline), day 5, and day 10 to analyze levels of glucose, triacylglycerol, cholesterol, and cortisol. On day 10, rats were euthanized and livers were collected to observe the severity of inflammation. The results indicated that rats receiving SHP 450 mg/kg and the mixture of pellet with SHP 450 mg/kg showed a similar ability as those receiving diazepam 0.18 mg/kg to cope with stress, indicated by an improvement in all blood biochemistry parameters. Supplementation with SHP 450 mg/kg can be used as an alternative source of functional food for overcoming oxidative stress, as indicated by its ability to improve levels of blood glucose, triacylglycerol, total cholesterol, and cortisol, and to improve liver histology by decreasing severity of liver inflammation.
Mi merupakan salah satu produk pangan yang populer dan disukai oleh berbagai kalangan masyarakat di Indonesia. Kandungan mi pada umumnya memiliki karbohidrat yang tinggi, rendah lemak, dan kandungan protein rendah. Untuk meningkatkan kadar protein dilakukan fortifikasi dengan penambahan ikan dalam bentuk tepung. Penelitan ini bertujuan untuk mengetahui tingkat kesukaan dan karakteristik fisik mi basah yang difortifikasi dengan jenis ikan yang berbeda. Penelitian ini menggunakan Rancangan Acak Lengkap dengan empat perlakuan yaitu tanpa penambahan ikan sebagai kontrol, penambahan tepung ikan lele, tepung ikan gurame dan tepung ikan patin. Konsentrasi tepung ikan yang ditambahkan yaitu 15%. Pengujian yang dilakukan pada mi basah meliputi uji organoleptik yaitu aroma, warna, tekstur dan rasa. Sedangkan uji fisik meliputi uji benda asing dan dayaserap air (DSA). Penambahan jenis tepung ikan yang berbeda berpengaruh nyata terhadap tingkat kesukaan aroma, warna, tekstur dan rasa mi basah yang dihasilkan. Pada uji fisik DSA (daya serap air) menunjukkan hasil yang berbeda nyata dibandingkan dengan kontrol. Pada uji fisik benda asing tidak menunjukkan adanya benda asing seperti pasir, tanah atau debu pada mi basah. Penambahan tepung ikan menghasilkan nilai DSA rata-rata 110%, sedangkan pada kontrol daya serap air pada mi ikan sebesar 80%.
Tujuan dari penelitian ini untuk menganalisis kandungan senyawa fenol dari alga cokelat Sargassum polycystum yang berasal dari Perairan Pantai Selatan, Gunung Kidul, Yogyakarta, secara kuantitatif terkait kandungan total fenolnya dan secara kualitatif yang meliputi uji Kromatografi Lapis Tipis dan HPLC (High Performance Liquid Chromatography). Polifenol diekstrak dengan etanol 96% dan dihitung rendemennya. Rendemen yang dihasilkan dari alga segar menjadi alga bubuk sebesar 9%, sedangkan rendemen dari alga segar menjadi ekstrak etanolik alga sebesar 5,1%. Nilai kandungan total fenol ekstrak pada konsentrasi 1000 ppm sebesar 59,30±0,58 g GAE/100 g ekstrak kering. Uji KLT dilakukan dengan standar asam galat dan floroglusinol, sedangkan uji HPLC dilakukan dengan standar asam galat, floroglusinol dan fenol. Hasil Uji KLT dan HPLC menunjukkan bahwa ekstrak etanolik dari Sargassum polycystum mengandung polifenol yang menyerupai standar asam galat dan fenol.
