Toxoplasmosis and neosporosis have been recognized as economically important diseases with considerable impact on the livestock industry. Considering the scarce information on the occurrence of Toxoplasma gondii and Neospora caninum infections in sheep from Uberlândia, Minas Gerais State, Brazil, this study aimed to investigate the frequency of antibodies against these parasites in sheep sera from this region by using different serological methods. A total of 155 sheep serum samples were analyzed by the indirect fluorescence antibody test (IFAT) and enzyme-linked immunosorbent assay (ELISA) for the detection of IgG against T. gondii and N. caninum. Seroreactivity by IFAT showed 80% of samples with titers between 512 and 2048 for T. gondii (cutoff ≥ 64) and 78% presenting titers between 50 and 200 for N. caninum (cutoff ≥ 50). Seroreactivity by ELISA showed 75% of samples with ELISA index (EI) between 2.0 and 3.0 for T. gondii (cutoff ≥ 1.3) and 54% presenting EI between 1.3 and 2.0 for N. caninum (cut off ≥ 1.3). Discordant results by both tests were analyzed by immunoblot, resulting in a total seropositivity of 61% for T. gondii and 23% for N. caninum, with 41% to T. gondii only, 3% to N. caninum only, and 20% to both parasites. There was a significant positive association between seropositivity to T. gondii and age over one year (P<0.001), but such association was not found for N. caninum infection. In conclusion, as T. gondii and N. caninum infections are simultaneously present in sheep flocks of this region, it should be emphasized the importance to carry out a regular monitoring of Toxoplasma infection due to its high prevalence, its zoonotic potential and induction of reproductive disorders leading to economic losses. For neosporosis, sheep farmers should be instructed about the presence of the parasite in the flock, its risk factors and potential abortifacient role in sheep. Differential flock management could be valuable tool to establish the association of serological positivity and reproductive disease induced by N. caninum in sheep.
ABSTRACT:Leptospirosis is a zoonotic disease that occurs worldwide, affecting pets and livestock that can be a source of infection for humans, considered therefore an important disease in public health. Thus, an investigation was made into the occurrence of anti-Leptospira agglutinins in several animal species on a farm located in the municipality of Uberlândia, state of Minas Gerais, Brazil. A total of 172 blood serum samples were collected from five species: cattle, dogs, goats, horses and sheep. The diagnosis was performed by means of the microscopic agglutination test (MAT), using a collection of 12 live antigens. Of the 172 animals evaluated, 62 (36%) were seroreactive, and the most frequent serovars in each species were: Hardjo (54.54%), Icterohaemorrhagiae (27.27%), and Autumnalis (18.18%) in cattle; Icterohaemorrhagiae (37.5%), Autumnalis (31%), Canicola (18.7%) and Hardjo (12.5%) in dogs; Grippotyphosa (75%) and Hardjo (25%) in goats; Icterohaemorrhagiae (50%), Grippotyphosa (37.5%) and Hardjo (12.5%) in horses, and Hardjo (34.78%); Grippotyphosa (26.08%); Bratislava (17.39%), Autumnalis (17.39%) and Icterohaemorrhagiae (4.4%) in sheep. It is believed that the occurrence of anti-Leptospira agglutinins can be traced back to a source of infection that is common to these species, or that the agent was probably transmitted between them because they live in close proximity to each other.
ABSTRACT:The aim of this study was to investigate the occurrence of antibodies against Leptospira spp. and risk factors related to the epidemiology of leptospirosis in dogs living in the urban area of the municipality of Uberlandia, Minas Gerais, Brazil. Blood serum samples were examined from 268 dogs from the municipality's North, South, East, West and Central sanitary districts. The samples were collected during an animal vaccination campaign against rabies in August 2008. The occurrence of antibodies against specific leptospiral serovars was determined by the microscopic agglutination test (MAT). Multivariable logistic regression was used to evaluate the association between sex, age, previous vaccination, habitation, presence of rats, and history of human and/or animal leptospirosis on the occurrence of infection. The occurrence of canine leptospirosis was 28.36% (76/268), the most prevalent serovars were: Autumnalis (34.21%), Tarassovi (23.58%), Canicola (17.11%) and Grippotyphosa (14.47%). No risk factors were found to be statistically significant (p> 0,05 for all examined variables). The fact that close to 30% of dogs showed evidence of past infection with Leptospira points to the need for the adoption of preventive measures, as the disease may represent a public health risk.
RESUMOO objetivo deste estudo foi determinar a soroprevalência de anticorpos anti-Neospora caninum e anti-Leptospira interrogans em vacas leiteiras do município de Goiandira, Goiás, Brasil. Testaram-se amostras de soro sanguíneo de 53 bovinos de dois rebanhos para detecção de anticorpos anti-N. caninum e anti-L. interrogans, utilizando as técnicas de Reação de Imunufluorescência Indireta (RIFI) e Reação de Soroaglutinação Microscópica (MAT), respectivamente. Consideraram-se positivos os soros reagentes a uma diluição de 1:50 para N. caninum e 1:100 para L. interrogans. Encontraram-se 35,9% (14 vacas) dos animais positivos para N. caninum na Fazenda 1, e 64,3% (9 vacas) dos animais da Fazenda 2. Para leptospirose, foram sororreagentes 87,2% (34 vacas) dos animais da Fazenda 1 e 64,3% (9 vacas) dos animais da Fazenda 2. Os sorovares encontrados foram: wolffi (21,74%), hardjo (18,11%); bratislava (14,49%); australis (12,32%); tarassovi (12,32%); icterohaemorrhagiae (7,25%); sentot (5,80%); grippotyphosa (5,07%); patoc (2,17%); cynoptero (0,72%). De acordo com esses resultados, é possível concluir que neosporose e leptospirose são doenças endêmicas em bovinos e se caracterizam como relevante problema de saúde pública e animal em Goiandira, GO, Brasil.
