RESUMOO presente trabalho apresenta um modelo de eletroforese que pode ser utilizado para o ensino de bioquímica proporcionando a compreensão desta técnica pelos estudantes. O objetivo desta eletroforese é oferecer o acesso a esta técnica com custos mais baixos do que o equipamento convencional. A fonte e a cuba foram construídas com componentes eletrônicos simples e de fácil acesso, com a mesma eficiência teórica da fonte e da cuba convencional. ABSTRACTThis paper presents a model of electrophoresis for biochemistry classes, were the comprehension of this technique is better absorbed by the students. The aim of this type of electrophoresis is to offer access to this technique with low costs than conventional equipment. The power and the tub were built with electronic parts simple and easy to access, with the same theoretical efficiency of conventional power and tub.
Resumo. Cynaeus angustus (LeConte) caracteriza-se como praga de grãos armazenados, possui diversos substratos alimentares listados em sua ecologia alimentar entre eles o milho, trigo, sorgo e soja. Porém não há registro de seus hábitos no amendoim in natura, assim o objetivo deste estudo foi apresentar a tabela de vida estática, para C. Angustus em condições de laboratório utilizando como substrato o amendoim, e avaliar a composição proteica nos diferentes estádios de desenvolvimento de C. angustus. A quantificação de proteínas foi feita pelo método de microbiureto e o método qualitativo foi feito per método eletroforético. Assim a quantificação de proteínas indicou um aumento de seus valores durante o desenvolvimento dos insetos: larvas 38,22 mg/mL, pupas 47,53 mg/mL e adultos de 60.00 mg/mL. Foi possível observar o perfil electroforético de C. angustus indicando um aumento significativo na variedade de proteínas (29 a 700 kDa) sobre as diferentes etapas deste inseto. Os dados obtidos pela analise populacional indicam que C. angustus apresenta maiores taxas iniciais de sobrevivência, alto potencial biótico e desenvolvimento contínuo em condições favoráveis que mostram potenciais condições de cultivo e pode ser usado como uma fonte nutricional tal como mencionado para outras espécies da mesma família.Protein Evaluation and Static life Table Cynaeus angustus (LeConte) (Coleoptera: Tenebrionidae) Abstract. Cynaeus angustus (LeConte) is characterized as a pest of stored grain, there is several food substrates on their feeding ecology including corn, wheat, sorghum and soybeans. But there is no record of their habits in peanut in natura, so the aim of this study was to present the static life table, to C. angustus in laboratory conditions using as substrate the peanut, and evaluate the protein composition in different stages of development C. angustus. The quantification of proteins was taken by microbiureto method and qualitative method was done per electrophoretic method. Protein quantitation indicated an increase of its values during development of insects: larvae 38,22 mg/mL, pupae 47,53 mg/mL and adults 60,00 mg/mL. It was possible to observe the electrophoretic profile C. angustus showed a significant increase in the variety of proteins (29-700 kDa) on the different stages of the insect. Data obtained from the population analysis indicate that C. angustus shows higher initial survival rates, high biotic potential and continuous developing in favorable conditions showing potential growing conditions and, that can be used as a nutritional source such as mentioned for other species of the same family.
ABSTRACT:The jau, Zungaro jahu (Ihering, 1898) is an endangered species, rheophilic, reaching 1.5 m and more than 100 kg. During a work of microbiological monitoring of water quality, in the cultivation of Zungaro jahu, microorganisms that were isolated, were previously reported in other studies, as family members Enterobacteriaceae, Staphylococcus and non-fermenting bacilli. However, it was also isolated Chromobacterium violaceum, which despite being a bacterium found in such an environment, it was reported linked to the cultivation of fish. This paper describes the Chromobacterium violaceum founded in a psiculture, which has implications for national fish farming.
Introduction: Candida albicans is the main agent of the most common fungal infection, Candidiasis. It is an opportunistic and dangerous pathogen, especially in immunosuppressed patients. The biological properties of Morinda citrifolia (noni) make it a potent antifungal. In this study, antifungal effect of M. citrifolia was evaluated to verify its effect on human cells. Methodology: Extract of M. citrifolia was used against strains of C. albicans (cEC 1291). Glucose consumption in C. albicans biofilm was determined at different concentrations of M. citrifolia, and germ tube formation was evaluated in the presence and absence of M. citrifolia. Fungicidal activity was determined by the kinetics of fungal cell death. THP-1 and HeLa cells were used for cell viability and apoptosis, and cell proliferation assays, respectively. Results: Cells treated with M. citrifolia maintained higher concentration of glucose than the control group (p < 0.05). Germ tube formation was inhibited in cells treated with M. citrifolia (p < 0.05). M. citrifolia exerted a cytotoxic effect on C. albicans cells with 99.99% lethality after 6.82 h (1:1 and 1:2), and reduced the viability of THP-1 cells by 25% and 67% after 12 and 36 h, respectively. Annexin V expression in THP-1 increased in groups that received higher concentrations of M. citrifolia (p < 0.05), reducing the proliferation of THP-1 and HeLa cells (2.8-fold). A greater cytotoxic effect was observed in fungal cells. Conclusions: These results indicate that M. citrifolia exerts biological activity against C. albicans and reduces the viability and proliferation of human cells.
Teaching of biology and chemistry in the schools has been widely discussed. The use of demonstration classes to content assimilation explained in lectures has been rarely used; hampering a better understanding of the students explained the content in the classroom. This article presents a proposal in science education combining theory and practice to facilitate the capture knowledge by students. The use of a simple distillation system for the explanation of metabolic pathways is a methodology that may present improvements in learning.
