The local duck DNA barcoding is still rarely conducted in Indonesia while DNA barcoding is extensively used as a tool of species identification and delineation tool. This study aimed to analyze the sequence and patterns of Central Javanese ducks mitochondrial cytochrome c oxidase subunit 1 (COI) gene. Feather samples of seven breeds of native duck were collected from traditional husbandries in Central Java. The samples were employed for DNA extraction and COI gene amplification. Five haplotypes were obtained from 35 samples, i.e., haplotype A, B, C, D, and E. Also, 9 variable sites with synonym substitution was detected in four nucleotides number 55, 61, 100, and 109; whereas five synonym substitutions were identified in the nucleotides number 36, 48, 51, 66, and 756. In conclusion, this study annotates that COI mtDNA gene is essential for local ducks barcoding system.
Tomato is one of the high antioxidant potential vegetables. Nowadays, there are many techniques of tomato processings instead of fresh consumption, i.e. boiled, steamed, juiced and sauteed. Every treatment of cooking will influence the chemical compound inside the fruits and the body's nutrition intake. It is important to conduct the research on antioxidant compound especially lycopene, β-carotene, vitamin C, α-tocopherol, and its activity after processing. This research has been done using the experimental method. Tomatoes were cooked into six difference ways, and then it was extracted using the same procedure continued with antioxidant measurement. The research results showed that steaming had promoted the higher antioxidant numbers (lycopene. α-tocopherol, β-carotene and vitamin C) and higher TCA and antioxidant activities in the tomatoes than other processings. It was indicated that steaming was the best way to enhance amount, capacity and activities of antioxidants of the tomatoes.
Biology is one of the cross-interest subject that much demanded by high school students who take a specialization group of Social and Language studies, but the study result of biology is relatively low. This research aims to determine the correlation between: (1) perception with study result of biology;(2) interest with study result of biology; and (3) perception and interest with study result of biology together. The research was conducted at SMAN 1 Ambarawa, population and sample are all 10th grade students of Cross-Interest Biology Class in the academic year 2016/2017. The research used non experimental quantitative design with correlational analysis. The data collected in the form of data of student's perception, interest, and study result of biology. Data of perception and interest obtained by instrument of psychological scale with likert model, whereas data of study result obtained from the student value of final exam in the odd semester. The research results showed that:(1) the value of rx1y=0,213 (low category) with the value of Sig. 0,014 < 0,05 at significance level of 5%; (2) the value of rx2y=0,159 (very low category) with the value of Sig. 0,070 > 0,05 at significance level of 5%; and the value of R=0,216 (low category) with the value of Sig. 0,048 < 0,05 at significance level of 5%. Based on the research can be concluded that: (1) there is low positive correlation between perception with study result of biology; (2) there is no correlation between interest with study result of biology; and (3) there is low positive correlation between perception and interest with study result of biology together for 10th grade students of Cross-Interest Biology Class at SMAN 1 Ambarawa. AbstrakBiologi adalah salah satu mata pelajaran lintas minat yang banyak dituntut oleh siswa SMA yang mengambil kelompok spesialisasi IPS dan Bahasa, tetapi hasil studi biologi relatif rendah. Penelitian ini bertujuan untuk mengetahui hubungan antara: (1) persepsi dengan hasil belajar biologi; (2) minat dengan hasil studi biologi; dan (3) persepsi dan minat dengan hasil studi biologi bersama. Penelitian ini dilakukan di SMAN 1 Ambarawa, populasi dan sampel adalah semua siswa kelas 10 dari Kelas Biologi Cross-Interest pada tahun akademik 2016/2017. Penelitian ini menggunakan desain kuantitatif non eksperimental dengan analisis korelasional. Data dikumpulkan dalam bentuk data persepsi siswa, minat, dan hasil belajar biologi. Data persepsi dan minat diperoleh dengan instrumen skala psikologi dengan model likert, sedangkan data hasil belajar diperoleh dari nilai siswa ujian akhir pada semester ganjil. Hasil penelitian menunjukkan bahwa: (1) nilai rx1y = 0,213 (kategori rendah) dengan nilai Sig. 0,014 <0,05 pada taraf signifikansi 5%; (2) nilai rx2y = 0,159 (kategori sangat rendah) dengan nilai Sig. 0,070> 0,05 pada taraf signifikansi 5%; dan nilai R = 0,216 (kategori rendah) dengan nilai Sig. 0,048 <0,05 pada taraf signifikansi 5%.
