Nanoceria (cerium dioxide nanoparticles, CeO2) has a broad range of biological properties including antiviral activity. The hypothesis was that nanoceria can efficacy against coronavirus (coronavirus of porcine transmissible gastroenteritis) and potentially can target SARS-CoV-2. Transmissible gastroenteritis coronavirus (TGEV) is the etiologic agent of porcine transmissible gastroenteritis (PTG), a highly contagious pig intestinal disease. The aim of the study was to determine the antiviral activity of CeO2 nanoparticles on the model of porcine coronavirus – TGEV. Methods. We used a highly pathogenic virus strain D52-5 (BRE79), of TGEV. We evaluated antiviral activity of CeO2 nanoparticles on the experimental model of porcine coronavirus (transmissible gastroenteritis virus) in transplantable line of porcine embryonic kidney cells (PEK) culture. Results. The criterion for evaluating the inhibitory activity of antiviral drugs in different in vitro systems is the selectivity index (SI) and the reduction of infectious titer by 1.5–2.0 lgTCD50. Nanoceria effectively inhibited the reproduction of porcine coronavirus with SI index of 83.3.
Objective. Determination of the virus-cidal and bacterio-cidal action of the ozone-containing steam-water mixture, which was obtained, using apparatus POS-1, simulated on viral models of transmissive gastroenteritis of pigs of the coronaviruses family and polyresistant clinical strains of bacterial cultures, as well as adjustment of optimal parameters of ozone in the gaseous mixture content, which do not cause pathological changes in the organism’s organs and systems. Materials and methods. Apparatus POS-1, created by collective of Scientific-Methodical Centre «Medical Innovation Technologies»», was applied for production of the ozone-containing gaseous mixture. Virusological investigations were conducted on the base of the Institute of Epidemiology and Infectious Diseases named after L. V, Gromashevskiy. As a working material the models of virus of the pigs transmissive gastroenteritis of the coronaviruses family were applied. The series of bacteriological and experimental investigations were conducted on the base of Shalimov National Institute of Surgery and Transplantology NAMS of Ukraine. Polyresistant cultures in concentration of 108 colony-creating units per 1 ml were applied for bacteriological investigations. The experiment was conducted on white rats to study the gaseous ozone-containing mixture affection on living biological tissues. Results. Virusological investigations have shown, that in the 20 minutes exposition and a contact with extracellular virus the infection titer have lowered in 100 000 times, and in a 30 minutes exposition - a complete deactivation of virus have had occurred. Bacteriological investigations have revealed the complete absence of the cultures development after their processing with the ozone-containing mixture during 20 min. The results of swimming tests and behavioral reactions in white rats of control and experimental groups did not differ. Histological investigations of the respiratory organs tissues as well as of spleen, thyroid gland, kidneys and suprarenal glands did not reveal pathological effects of the ozone-containing mixture. Conclusion. Parameters of the ozone generation and delivery in the gaseous mixture content were studied and optimally selected. Investigations of the ozone-containing mixture affection on a viral strain of the coronaviruses family, as well as bacteriological investigations on polyresistant bacterial cultures have had confirmed its pronounced virus-cidal and bactericidal properties. Application of the ozone-containing mixture do not cause any pathological changes in the living organism organs and systems. This gaseous mixture may be used for prevention and treatment of respiratory infectious diseases of viral and bacterial genesis.
Relevance. Seasonal differences in stroke incidence are associated with various physical factors, some associated with pathophysiological changes in the body leading to ischemic stroke. At the same time, there is little research into the differences in the frequency of detection of the genome of herpes virus infection and influenza virus, depending on the season. Objective. Investigate the existence of associations between the season and the frequency of virologically confirmed herpezoid infection manifested by herpezoid virus and influenza virus of patients with ischemic stroke. Materials and methods. In the period from 01.01.2017 to 31.12.2017. during the year at the bases of the neurological and department of the Alexandrovsky Clinical Hospital Centre. Kyiv conducted a study of 144 cases of cerebral ischemic stroke with the medium-severe neurological deficit: 78 (54.2%) women and 66 (45.8%) men, with an average age of 63.1 0.8 years. Neurological examination and identification of the genome of herpes viruses and influenza virus, performed with the help of a polymerase chain reaction (PCR), was carried out monthly in 12 patients hospitalized by ambulance. The account was taken of the presence of a viral manifestation if it preceded the stroke within 2 weeks. Results. 36 cases were examined each season. The ratio of men to women was 45.8 / 54.2 in total and did not differ between seasons, p = 0.514. 32 (22.2 percent) patients have demonstrated a viral infection. In winter, 11 (34.4 percent), in spring, 7 (21.9 percent), in summer, 4 (12.5 percent), in autumn, 10 (31.3 percent), and all p. At the same time, during the summer period, the incidence of viral manifestation was definitely lower than in winter, p = 0.042. Virus genomes were found in 12 (33.3 percent) patients in winter, 7 (19.4 percent) in spring, 5 (13.9 percent) in summer, 12 (33.3 percent) in autumn and p = 0.131 rice. During the summer period, the frequency of PCR-confirmed herpes virus infection was definitely lower than in the winter-autumn season, p = 0.033. Conclusions. Viral manifestations of herpes and influenza virus in the blood are more frequent during the winter and autumn periods; the frequency of detection indicated viral infection from October to January is reliably higher than the rate from March to August inclusive. In 25.0 percent of patients with ischemic stroke, the genome of herpes viruses and influenza virus in the blood is detected. In the event of a demonstration, the frequency of the above-mentioned viral infection is clearly higher (90.6 percent as against 9.4 percent) than that of patients without it.
