We investigated the effect of elevated [CO2], [O3] and temperature on plant productivity and if these climate factors interacted with each other in multifactor treatments. The climate effects were studied in 14 different cultivars/lines of European spring oilseed rape (Brassica napus L.) and spring barley (Hordeum vulgare L.). Seven genotypes of each species were cultivated in six single‐ and multifactor treatments with ambient or elevated CO2 (385 ppm and 700 ppm), O3 (20 ppb and 60 ppb) and temperature (12/19 °C and 17/24 °C). Growth and production parameters were measured. Elevated CO2 increased yield and biomass. Seed number increased by about 47 % in barley and by 26 % in oilseed rape, but in oilseed rape, the TSW was significantly decreased, possibly because of shortening of the seed filling period. Higher temperatures decreased yield and biomass significantly in both species. A significantly decreased yield and thousand grain weight was also seen in barley due to elevated O3. The multifactor combination of elevated CO2, O3 and temperature showed a decrease in growth and production in the two species, though not statistically significant for all parameters. This trend suggests that the expected increase in the plant production in northern Europe, indicated by Intergovernmental Panel on Climate Change (IPCC) as a consequence of increased [CO2] and temperature, may not hold, due to interactions between these abiotic factors.
Black carrots are characterized by a significant amount of anthocyanins, which are not only a good source of natural food colorant, but can also provide many health benefits to humans. In the present work, taproots of different carrot genotypes were used to identify the candidate genes related to anthocyanin synthesis, with particular a focus on R2R3MYB, bHLH transcription factors, and glutathione S-transferase gene (GST). The RNA-sequencing analysis (RNA-Seq) showed that DcMYB6 and DcMYB7 had a genotypic dependent expression and they are likely involved in the regulation of anthocyanin biosynthesis. They were specifically upregulated in solid black taproots, including both black phloem and xylem. DcbHLH3 (LOC108204485) was upregulated in all black samples compared with the orange ones. We also found that GST1 (LOC108205254) might be an important anthocyanin transporter, and its upregulated expression resulted in the increasing of vacuolar anthocyanin accumulation in black samples. Moreover, high performance liquid chromatographic (HPLC) analysis and liquid chromatography coupled to mass spectrometry (LC-MS) were used to identify the individual anthocyanin in the purple tissues of two carrot cultivars. The results showed that five main anthocyanin compounds and the most abundant anthocyanin were the same in different tissues, while the second-highest anthocyanin between three tissues was different, even in the same cultivar. In conclusion, this study combined anthocyanin profiles and comparative transcriptomic analysis to identify candidate genes involved in anthocyanin biosynthesis in carrots, thus providing a better foundation for improving anthocyanin accumulation in carrots as a source of colorants.
The portion-dilution model is a parametric restatement of the conventional view of animal pollination; it predicts the level of pollinator-mediated gene dispersal. In this study, the model was applied to white clover (Trifolium repens) and its most frequent pollinator, the honeybee (Apis mellifera). One of the three parameters in the portion-dilution model is the mean number of flowers a pollinator visits in one foraging bout. An alternative method to estimate this parameter was developed that was not depending on pollinator hive-seeking behaviour. The new estimation method, based on nectar collection, seems to be a good alternative, when reliable observation on visiting behaviour of pollinators is not possible. The gene flow in white clover was modelled. Where fields were assumed to be well separated, and only a low fraction of bees travelled between fields, the gene flow was estimated to be 0.7%, but subjected to large uncertainty. In a worst case scenario with adjacent fields-one with a genetically modified (GM) T. repens cultivar and the other with a conventional T. repens cultivar-and where all arriving bees were expected to transfer GM pollen, the median gene flow was modelled to be 7% with an estimated 95% percentile of 70%. The results show that the European Union threshold limit of 0.9% GM admixture for food and feed will likely be exceeded at times and especially organic farmers that do not accept GM admixture and often have clover and clover-grass fields might face challenges with admixture of GM.
