Cancer cells display phenotypic equilibrium between the stem-like and differentiated states during neoplastic homeostasis. The functional and mechanistic implications of this subpopulation plasticity remain largely unknown. Herein, it is demonstrated that the breast cancer stem cell (BCSC) secretome autonomously compresses the stem cell population. Co-implantation with BCSCs decreases the tumor-initiating capacity yet increases metastasis of accompanying cancer cells, wherein DKK1 is identified as a pivotal factor secreted by BCSCs for such functions. DKK1-promotes differentiation is indispensable for disseminated tumor cell metastatic outgrowth. In contrast, DKK1 inhibitors substantially relieve the metastatic burden by restraining metastatic cells in the dormant state. DKK1 increases the expression of SLC7A11 to protect metastasizing cancer cells from lipid peroxidation and ferroptosis. Combined treatment with a ferroptosis inducer and a DKK1 inhibitor exhibits synergistic effects in diminishing metastasis. Hence, this study deciphers the contribution of CSC-regulated phenotypic plasticity in metastatic colonization and provides therapeutic approaches to limit metastatic outgrowth.
Patterns of precipitation have changed as a result of climate change and will potentially keep changing in the future. Therefore, it is critical to understand how ecosystem processes will respond to the variation of precipitation. However, compared to aboveground processes, the effects of precipitation change on soil microorganisms remain poorly understood. Changbai Mountain is an ideal area to study the responses of temperate forests to the variations in precipitation. In this study, we conducted a manipulation experiment to simulation variation of precipitation in the virgin, broad-leaved Korean pine mixed forest in Changbai Mountain. Plots were designed to increase precipitation by 30 % [increased (+)] or decrease precipitation by 30 % [decreased (-)]. We analyzed differences in the diversity of the bacterial community in surface bulk soils (0-5 and 5-10 cm) and rhizosphere soils between precipitation treatments, including control. Bacteria were identified using the high-throughput 454 sequencing method. We obtained a total 271,496 optimized sequences, with a mean value of 33,242 (±1,412.39) sequences for each soil sample. Being the same among the sample plots with different precipitation levels, the dominant bacterial communities were Proteobacteria, Acidobacteria, Actinobacteria, Planctomycetes, and Chloroflexi. Bacterial diversity and abundance declined with increasing soil depth. In the bulk soil of 0-5 cm, the bacterial diversity and abundance was the highest in the control plots and the lowest in plots with reduced precipitation. However, in the soil of 5-10 cm, the diversity and abundance of bacteria was the highest in the plots of increased precipitation and the lowest in the control plots. Bacterial diversity and abundance in rhizosphere soils decreased with increased precipitation. This result implies that variation in precipitation did not change the composition of the dominant bacterial communities but affected bacterial abundance and the response patterns of the dominant communities to variation in precipitation.
With the rapid development of computer technology, BIM has become the most common tool in the construction industry, which occupies an important position in the virtual construction of Prefabricated building (hereinafter referred to as PB). Through the establishment of virtual construction engineering model, we can provide a complete construction engineering information base combined with digital technology. In the virtual construction of construction engineering, we can check the collision through BIM. At the same time, through BIM, we can carry out 3D model drilling of virtual construction. Through collaborative management, we can improve the production efficiency, which will save the cost and shorten the construction period. At present, the application of BIM in China is still in the exploratory stage, which requires us to give full play to BIM Technology. Through the Revit software, this paper simulates the construction site, buildings, tower crane, secondary masonry, etc., which can make the application of PB more clear. Firstly, this paper analyzes the related concepts. Then, this paper analyzes the application of BIM in the virtual construction of PB. Finally, some suggestions are put forward.
N6-methyladenosinen (m6A) methylation is a frequent RNA methylation modification that is regulated by three proteins: “writers”, “erasers”, and “readers”. The m6A modification regulates RNA stability and other mechanisms, including translation, cleavage, and degradation. Interestingly, recent research has linked m6A RNA modification to the occurrence and development of cancers, such as hepatocellular carcinoma and non-small cell lung cancer. This review summarizes the regulatory role of m6A RNA modification in gastric cancer (GC), including targets, the mechanisms of action, and the potential signaling pathways. Our present findings can facilitate our understanding of the significance of m6A RNA modification in GC.
