Early life is a decisive stage for the development of physiological and psychological characteristics of an individual. Any stress or disruption of healthy development at this stage has serious long-lasting consequences for the remaining life. Unfortunately, early life stress is a common occurrence in humans and other animals. In this context, we investigated if the provision of environmental enrichment during the pre-weaning phase of rat pups and dams could alter the consequences of early-life maternal-separation stress. Pre-weaning enrichment rescued the effects of maternal separation on the excess secretion of adrenal stress hormones and anxiety-like behavior during adulthood. Enrichment also reduced the effect of stress on the spine density of basolateral amygdala neurons, a brain region critical for stress-induced facilitation of emotional behaviors. Pre-weaning enrichment, provided during early-life, blunted the effects of maternal separation stress on decreased intra-nuclear translocation of glucocorticoid receptors within the amygdala neurons when tested later in adulthood. Early-life, pre-weaning environmental enrichment also increased the amount of brain-derived neurotrophic factor within adult basolateral amygdala. Our observations showed that environmental manipulation during early formative years could be utilized to build lifelong resilience to stress. Complex naturalistic housing and sensory enrichment is, thus, an useful buffer against an impoverished and stressful childhood.
Selective serotonin reuptake inhibitors (SSRIs) are the most commonly used medications for mood and anxiety disorders, and adult neurogenesis in the dentate gyrus has been shown to be involved in the behavioral effects of SSRIs in mice. Studies have shown the varied effects of chronic treatment with SSRIs on adult neurogenesis. One such effect is the acceleration of neuronal maturation, which affects the functional integration of new neurons into existing neuronal circuitry. In this study, we labeled new neurons by using GFP-expressing retroviral vectors in mice and investigated the effect of an SSRI, fluoxetine, on these neurons at different time points after neuronal birth. Chronic treatment with fluoxetine accelerated the dendritic development of the newborn neurons and shifted the timing of the expression of the maturational marker proteins, doublecortin and calbindin. This accelerated maturation was observed even after sub-chronic treatment, only when fluoxetine was administered during the second week of neuronal birth. These results suggest the existence of a ‘critical period’ for the fluoxetine-induced maturation of new neurons. We propose that the modified functional integration of new neurons in the critical period may underlie the behavioral effects of fluoxetine by regulating anxiety-related decision-making processes.
We describe a pilot community-based conservation initiative for wolves Canis lupus that involves (i) voluntary deactivation of traditional trapping pits called Shandong, (ii) commitment to wildlife conservation by the local community, and (iii) collaborative construction and consecration of a Stupa (Buddhist shrine) in the vicinity of the Shandong as a symbol of conservation and repentance for past hunting. People and wolves have a complex relationship, in part shaped by predation on livestock, which can have severe impacts on livelihoods in pastoral societies. Consequently, wolf conservation often evokes strong and polarizing reactions. To control wolf populations, livestock herders across the Trans- Himalayan and Tibetan regions use different types of traps. Shandong is a relatively large, widely used traditional trapping pit with inverted funnel-shaped stone walls, usually built near villages or herder camps. Typically, a live domestic animal is placed in the pit to attract the wolves. Once the wolves jump into the pit, the funnel shaped walls prevent them from escaping, and trapped wolves are usually stoned to death. In an extensive survey covering over 25,000 sq. km, we enumerated 94 Shandong in 58 of the 64 surveyed villages in Ladakh between June 2019 and March 2020. Thirty of these had been used to kill wolves within the past 10 years, while 7 had been destroyed. Shandong that were not in use were of poorer condition. Since 2017, we have worked with community members, local monks, and the region’s religious leaders to support the neutralization of the Shandong while preserving their structure, and assisted the communities to build Stupas and to consecrate them. Our pilot efforts with three communities appear to generate pride locally, and hold promise for promoting wolf conservation in Ladakh and in large parts of Trans-Himalayan and Tibetan regions that share similar cultural settings.
