A survey to investigate the stem and bulb plant parasitic nematode at one of garlic area centre, in Temanggung, Central Java, Indonesia was conducted from Januaryto February 2018. Infected plant with specific symptoms, morphological and morphometric characters both of female and male of adult nematodes were used to describe a A1 quarantine plant parasitic nematode Ditylenchus dipsaci. Seven from nine observed locations were postively infected with population in average 2.67 nematodes per 100 g of soil and 2.67–189.33 per bulb. This is the first report of D. dipsaci from Indonesia and consequently further investigations were needed to know their distribution and also to confirm the origin of the nematode. Tanaman terinfeksi dengan gejala serangan yang spesifik, karakter morfologi dan morfometri nematoda betina dan jantan dewasa dipergunakan dalam identifikasi Ditylenchus dipsaci yang merupakan nematoda parasit tanaman yang termasuk OPTK A1 (Organisme Pengganggu Tanaman Karantina A1) di Indonesia. Sebanyak tujuh dari sembilan lokasi yang diamati mengindikasikan positif sebagai daerah sebaran nematoda tersebut dengan populasi rata-rata 2,67 ekor per 100 gram tanah dan 2,67–189,33 nematoda per umbi. Laporan pertama tentang nematoda D. dipsaci ini membawa konsekuensi perlunya penelitian lebih lanjut untuk mengetahui daerah sebaran dan juga konfirmasi dari mana nematoda tersebut berasal.

Root-knot nematodes are considered as one of the most destructive pathogens of potatoes, especially on tuber seed. Infected potato seed will cause tuber malformation and the most important thing is as the main spreading source of Meloidogyne spp. The objective of this research was to know the prevalence and identify the species of root-knot nematode which attack the potato seed in four sub-districts of potato production centre in Central Java, Indonesia. Molecular and morphological identification was conducted for the nematode species identification. PCR assay using MIG primer to detect three tropical root-knot nematodes followed by sequencing was conducted for molecular detection, while the perennial pattern was conducted for morphological detection. Results showed that root-knot nematodes on potato seed were generally distributed in Central Java with the prevalence percentage between 14.28-88.23% on the three from four sampling area. The molecular and morphological identification show that species of root-knot nematodes identified on potato seeds were Meloidogyne javanica, M. incognita, and M. arenaria.
The demand for daily consumption of tomato fruit is increasing immensely. Nevertheless, the fruit is exposed to mechanical damage, shrinking, and softening as the maturity stages, handling, and storage are inappropriate, thereby affecting the texture. The study aimed to assess the texture profile, pectinase activity, and physicochemical parameters in tomato fruit at different maturity stages and storage temperatures. The fruits were harvested at 1-4 weeks after pollination and stored at a temperature of 16 ºC and 25ºC. There was an increase in the redness color (a*), TSS content, weight loss, respiration rate, and ethylene production, while the hardness, lightness color (L*), pH, and TA decreased with an increase in maturity stages at different storage temperatures. The higher Polygalacturonase (PG) and Pectin methylesterase (PME) enzyme activities were observed at 25 ºC compared to storage temperature of 16 ºC. It was confirmed that pectinase activity extremely affected the texture profile. For commercial purposes, it is suggested that tomatoes are harvested at 2nd and 3rd week after pollination for long distance transportation and at 4th week for fresh consumption and stored at a temperature of 16 °C.
