Ferroptosis is a new type of cell death marked by iron and lipid ROS accumulation. GPX4 is one of the glutathione peroxidases known to regulate ferroptosis tightly. On the other hand, Nrf2 also plays a vital role in ferroptosis as it targets genes related to oxidant defense. Herein, we employed beas-2 human epithelial cells treated with a low concentration of RSL3 to induce ferroptosis. To study the protective role of Nrf2, we used ML385 as its specific inhibitor. A combination of ML385 and a low concentration of RSL3 synergistically induced more toxicity to RSL3. Furthermore, we found that mitochondrial ROS is elevated in ML385 and RSL3 combination group. In addition, Mito TEMPOL application successfully prevents the upregulation of mitochondrial ROS, lipid ROS, reduces the toxicity of RSL3, restores the antioxidant capacity of the cells, and mitochondrial functions reflected by mitochondrial membrane potential and mitochondrial oxidative phosphorylation system (OXPHOS) expression. Altogether, our study demonstrated that Nrf2 inhibition by ML385 induces more toxicity when combined with RSL3 through the elevation of mitochondrial ROS and disruption of mitochondrial function.
Kondisi pandemi mempengaruhi berbagai aspek kehidupan manusia, termasuk aspek ekonomi. Pengelolaan keuangan yang kurang baik dapat menyebabkan ketidak seimbangan neraca keuangan rumah tangga. Kondisi pandemi menyebabkan masyarakat rentan sakit dan membutuhkan biaya perawatan. Mencegah sakit dapat dilakukan dengan memperbaiki pola hidup. Tujuan pengabdian masyarakat ini untuk memberikan pelatihan Ibu-Ibu PKK RW 09 Kwarasan,Nogotirto, Gamping Sleman mengelola keuangan rumah tangga dan menjaga kesehatan keluarga menuju keluarga tangguh di era pandemi. Metode kegiatan terdiri dari pelatihan pengelolaan keuangan dan evaluasi keuangan, pendampingan analisa keuangan rumah tangga dan strategi-strategi meningkatkan pendapatan keluarga. Selain itu, pada kegiatan pengabdian masyarakat ini juga diberikan sosialisasi bagaimana menjaga kesehatan di era pandemi dan upaya meningkatkan religiusitas. Hal ini akan mengurangi tingkat stress dan tetap sehat.
BACKGROUND: Dioscorea alata L. tubers (DA) are suspected to prevent diabetes mellitus (DM) because they have a low glycemic index. However, only a few reports about the anti-diabetic effect of DA were reported up to date. This study aims to analyze the effect of DA consumption on several diabetic biomarkers through in vitro, in vivo, and in silico analysis.METHODS: In vitro experiments were conducted by observing the anti-inflammatory activity of DA extract, steroidal saponins (SDA) isolated from DA, and diosgenin in lymphocyte cell cultures. The tumor necrosis factor (TNF)-α and interferon (IFN)-γ percentages were analyzed by flow cytometry. In vivo study involved 24 healthy adolescents that were given a boiled DA 10 hours post-prandial. The blood sugar levels were measured at 0, 30, 60, and 120 min after treatment. Furthermore, the in silico study was carried out by analyzing the active compounds and predicting the biological activity, the target proteins, and interactions of target proteins with compounds contained in DA .RESULTS: DA extract, SDA isolated from DA, and diosgenin at 50 µg/mL significantly reduced pro-inflammatory cytokines TNF-α and IFN-γ in lymphocyte cell culture. The blood glucose levels in the DA group were lower at 30 and 60 min after treatment. Based on the in silico study, the anti-diabetic activity of DA was speculated to be attributed to the mechanisms of anti-hyperglycemia, prevention of mitochondrial dysfunction, anti-inflammation, and treated insulin resistance. Several proteins included in the DM pathway became the protein target of compounds contained in DA.CONCLUSION: DA potentially have an anti-diabetic activity through several mechanisms.KEYWORDS: hyperglycemia, inflammation, insulin resistance, yam
Lansia di RW 4, Pakuncen adalah kelompok rentan Covid-19 sehingga diharapkan tetap tinggal di rumah pada masa pandemi ini. Lansia juga rentan mengalami permasalahan kesehatan kulit. Sebagian lansia mengalami gatal, meskipun tidak sampai menyebabkan pergi ke fasilitas kesehatan. Pemeriksaan untuk mendeteksi penyakit sistemik yang berhubungan dengan keluhan gatal, seperti kadar gula darah untuk deteksi diabetes, kolesterol dan asam urat perlu dilakukan secara rutin. Pemeriksaan ini dilakukan saat kegiatan posyandu lansia oleh kader kesehatan. Oleh karena itu perlu dilakukan kegiatan untuk meningkatkan pengetahuan kader kesehatan tentang perawatan kulit dan ketrampilan pemeriksaan darah untuk mendeteksi penyakit sistemik. Kader akan meneruskan pengetahuan dan keterampilan tersebut kepada lansia, Kegiatan pengabdian dilakukan dengan ceramah dan diskusi tentang perawatan kulit, dan pelatihan pemeriksaan kadar gula kolesterol dan asam urat di darah tepi. Ceramah dan diskusi dihadiri oleh 20 orang kader dan perwakilan lansia. Pelatihan pemeriksaan darah diiikuti oleh 5 orang kader kesehatan. Kegiatan ini telah menghasilkan kader yang terampil memeriksa darah kepada teman kader dan perwakilan lansia. Terdapat 83% lansia dan kader yang mempunyai pengetahuan baik tentang kesehatan dan perawatan kulit. Masyarakat menyatakan kegiatan ini dapat dikembangkan karena sesuai dengan kebutuhan masyarakat Pakuncen.
