Superoxide dismutase (SOD; EC 1.15.1.1), a high molecular weight component of the antioxidant defense system, provided promising results in the treatment of oxidative damage. Thermothrix sp., isolated from thermal spa water in Serbia, showed high superoxide dismutase activity. The SOD, from cell free extract, was purified to homogenity by ammonium sulfate precipitation, Sephadex G 75 gel filtration chromatography and QAE Sephadex ion exchange chromatography. The specific activity of the purified enzyme was 9191 U/mg. The purified enzyme was analyzed and partially characterized. SOD was localized in polyacrylamide gel by activity staining, based on the reduction of nitroblue tetrazolium (NBT) by superoxide. The enzyme molecular weight determined by gel chromatography is 37 kD. According to SDS PAGE it is composed of two subunits of equal size, joined by noncovalent interactions. The isoelectric point, assessed by isoelectric focusing is 5.3. The optimum pH for enzyme activity was in the range of 8 to 10. The optimum temperature for SOD activity was 60 ?C. After one hour of incubation at 40, 50 and 60 ?C the SOD activity increases, but at 80 ?C, the SOD is denaturated. After 24 hours of incubation at 25 ?C SO Dactivity only slightly decreases.
The aim of this study was the partial characterization of the antimicrobial peptide bacteriocin G 2 produced by probiotic bacteria Lactobacillus plantarum G 2 , which was isolated from a clinical sample of a healthy person. Antimicrobial substance was secreted in the supernatant of an L. plantarum G 2 culture, and showed a diverse spectrum of antimicrobial activity of all the tested strains of the genera Lactobacillus and the pathogenic bacteria Staphylococcus aureus and Salmonella аbony. Isoelectric focusing revealed that bacteriocin G 2 is a cationic peptide (pI about 10) with a molecular mass of 2.2 kDa according to tricine-sodium dodecyl sulphate-polyacrylamide gel electrophoresis, SDS-PAGE. The antimicrobial activity of bacteriocin G 2 was diminished by the proteolytic action of trypsin and proteinase K. Bacteriocin G 2 preserved its biological activity in the temperature range 40-60 °C (15 min), which was lost at 80 °C. Bacteriocin G 2 was stable in the pH range 2-9, while treatment with 1 % Tween 80 and 1 % urea resulted in increased antimicrobial activity. The probiotic strain L. plantarum G 2 produces the antimicrobial substance proteinaceous in nature with bacteriocin characteristics. Bacteriocin production is one of the key properties of probiotic bacteria with clinical potential as antiinfective agents, which will increase the likelihood of its in vivo efficacy.
Sažetak: U radu je analiziran kulturološki i poetički aspekt smrti odnosno skoro potpunog nestanka iz Dalmacije etničkih Italijana, pripadnika srednje i više građanske klase, u Desničinom romanu Zimsko ljetovanje (1950) i, komparativno, u romanu Dalmacijo stara Dalmacijo (1979) Ivana Katušića. Motiv smrti, ratne opasnosti i neposrednog ratnog okruženja predstavljaju istorijske okolnosti Drugog svetskog rata u Zadru, njegovom zaleđu, i u drugim dalmatinskim gradovima. Prva seoba građanskog stanovništva pod velom preteće smrti predstavlja pomeranje u zaleđe, koje predstavlja drugu, ruralnu sredinu. Strah od smrti u građanima iz Zadra podstiče složena antropološka, kulturološka i verska pitanja. Analizom likova pojedinih građana i seljaka ukazuje se na susret i sukob različitih kultura i civilizacija. Smrt će za građane Zadra biti dvostrukog karaktera-prvo seoba i potom konačno iseljavanje. Zimsko ljetovanje najavljuje, mada sa skepsom, klasni prevrat, koji će biti osnova novog socijalističkog uređenja na prostorima Jugoslavije, pa će rad ukazati i na smenu društvenih uređenja koja će se odvijati tokom Drugog svetskog rata i konačan epilog dobiti upravo krajem rata. 1 Ključne reči: egzil, kontrapunkt, identitet, mentalitet, društvene promene, građanska klasa, urbano, ruralno U opusu Vladana Desnice Dalmacija je sveprisutna zemlja dvojnosti. Nijedan pisac sa prostora Dalmacije nije u modernom periodu bolje razumeo, tumačio i umetnički izatkao antipodnu sliku ovoga regiona. Vladan Desnica je to mogao svojim svestranim obrazovanjem, životnim iskustvom čoveka koji je poreklom iz zaleđa, odrastao u Zadru i Splitu, poznavanjem mentaliteta Vlaja, srpskog i hrvatskog stanovništva u Zagori, ali i visokog građanstva slovenskog i romanskog porekla u urbanim sredinama Zadra i Splita. Zimsko ljetovanje je i taj oksimoronski spoj dvojne Dalmacije, paradoksalnih i bizarnih situacija u kojima su život i smrt dva lica jednog prostora i vremena. W utworach tych starzenie się ma konotację wyraźnie negatywną, gdyż osoby sędziwe przedstawione zostały tu w sposób antagonistyczny w stosunku do osób młodych.
