Background:Diabetes is a metabolic disease prevalent worldwide in all age group of people. The source of diabetes is due to an oxidation process that can produce free radicals. An increase in oxidative free radicals in the body is reported to be one of the several causes of diabetes. The best remedy to combat oxidative stress is the use of antioxidants, which inhibit and scavenge free radicals.Aim:This study has been undertaken to evaluate the antioxidant activity and antidiabetic effect of mulberry leaf extract in diabetic mice.Materials and Methods:Antioxidant activity of mulberry leaves was determined by 2,2-diphenyl-1-picryl-hydrazyl (DPPH) and ferric reducing/antioxidant power (FRAP) assay. Antidiabetic assay of mulberry leaf extract was analyzed by oral administration of leaf extract up to 3 weeks in diabetic mice induced by streptozotocin.Results:In vitro antioxidant activity in both DPPH and FRAP assays showed significantly (P < 0.05) higher inhibition of free radicals than that with ascorbic acid. Diabetic mice fed with mulberry leaf extract showed increment (+25.88%) in body weight and a significant reduction in blood glucose concentration (−71.58%). Further, glucose-6-phosphate dehydrogenase enzyme activity was significantly (P < 0.05) increased, whereas activities of other enzymes particularly catalase, serum glutamic oxaloacetic transaminase, serum glutamic pyruvic transaminase were decreased in diabetic mice after oral administration of mulberry leaf extracts. Histology of liver revealed regeneration of hepatocytes, central vein, and nucleus.Conclusion:This study demonstrated that S-1708 mulberry variety has a potential therapeutic value in diabetes and related complications.SUMMARY Diabetes mellitus is a grave metabolic deviations and responsible for many complications affecting various organs in the human body. In spite of the known antidiabetic medicine available in the market, diabetes and the associated impediments sustained to be a major medical crisis. Medicinal plants have been proven to be useful in diabetes due to their rich therapeutic value. In the current study, S-1708 mulberry variety not only authenticated the earlier results obtained from other medicinal plants but also turn out to be known as a potential source for treating diabetes by demonstrating tremendous ant- diabetic properties. A simple and sensitive RP-HPLC method developed for estimation of Polmacoxib in bulk by using a high performance liquid chromatography with PDA detector using Phenomenex luna C18 column (250mmx4.6mm), 5 µm; mobile phase comprises of Water : ACN as (1:1) at the flow rate 1.0 ml/min and the wavelength of detection 238 nm and eluted at 8.12 minutes. The proposed method is validated for specificity, Linearity, Precision, accuracy, ruggedness, and Robustness. All the parameters were found within the acceptable limits. RP-HPLC method was a simple, reliable economic and acceptable and it confirmed that method is suitable for the intended use for routine quality control and assay of drugs. This method is successfully applied for the determination of commercial dosage form capsule preparation. This method is validated as per ICH (International conference on harmonization) Guidelines. Yograj guggulu vati (YGV), a polyherbal formulation is recommended for the management of diseases like arthritic, anodyne or analgesic, spasm, muscle relaxant, flatulence, digestive problem, cough, hyperglycaemia, fat burner and obesity. Though Yograj guggulu vati is widely used for the treatment of diseases in Ayurvedic System of Indian Medicine, but till date, it’s Phenolic and flavonoids contents and contamination studies have not been carried. In the present article, we evaluated the total phenolic and flavonoids contents and contamination of YGV. Total phenolic contents were evaluated by Folin Ciocalteu reagent. Aluminum chloride colorimetric method was used for the determination of total flavonoid contents. Contamination study such as microbial load was also performed. Microbial load study revealed that total bacterial counts and total fungal counts were under limits. The total phenolic content and total flavonoid content were 190.16 mg/g and 20.87 mg/ g dry extract respectively. Microbial load studies showed that the formulation has a good quality and purity. Presence of abundance phenolic and flavonoids compound indicated that YGV can be used for different biological activities. Keywords: Microbial load, Yograj Guggulu Vati, total phenolic contents
Psoriasis is a multi-factorial disease represented by complex atomic scenes and cellular pathways that lead to abnormal cell development. The normal mixture has a clear target and limited cytotoxicity; in this sense, it can support the further development of new therapies for the treatment of this flexible disease. Berberine is an individual of the original berberine alkaloid family. It mainly exists in the roots, trunks and barks of various trees and has anti-psoriatic activity. In any case, according to the berberine organization, limited bioavailability and low assimilation rate are the two main obstacles, because only 0.5% of the ingested berberine is consumed in the small digestive tract, and this rate is when it enters the intestines. It is further reduced to 0.35%. Nano-based details are seen as the best way to increase the assimilation rate because substances accumulated at the nano-level can be quickly absorbed in the intestine. Useful methods based on nanotechnology have been developed to solve these problems to ultimately achieve greater sufficiency in the treatment of various diseases. The investigation introduced and essentially examined the anti-proliferative effects of berberine and nanotechnology-based repair technology for nano-scale transportation of berberine. Finally, we will thoroughly analyze and examine the current methods and promising viewpoints of the recent transportation of this alkaloid.
