Silymarin is a flavonoid compound derived from milk thistle (Silybum marianum) seeds which has several pharmacological applications. Chalcone synthase (CHS) is a key enzyme in the biosynthesis of flavonoids; thereby, the identification of CHS encoding genes in milk thistle plant can be of great importance. In the current research, fragments of CHS genes were amplified using degenerate primers based on the conserved parts of Asteraceae CHS genes, and then cloned and sequenced. Analysis of the resultant nucleotide and deduced amino acid sequences led to the identification of two different members of CHS gene family,SmCHS1 and SmCHS2. Third member, full-length cDNA (SmCHS3) was isolated by rapid amplification of cDNA ends (RACE), whose open reading frame contained 1239 bp including exon 1 (190 bp) and exon 2 (1049 bp), encoding 63 and 349 amino acids, respectively. In silico analysis of SmCHS3 sequence contains all the conserved CHS sites and shares high homology with CHS proteins from other plants.Real-time PCR analysis indicated that SmCHS1 and SmCHS3 had the highest transcript level in petals in the early flowering stage and in the stem of five upper leaves, followed by five upper leaves in the mid-flowering stage which are most probably involved in anthocyanin and silymarin biosynthesis.
This study examined the influence of drought stress during the flowering stage of 2 canola cultivars on some physiological characteristics, antioxidant activities, and gene expression of antioxidant enzymes. Significant differences were observed among interactions of cultivars and levels of drought stress in total shoot dry material, relative water content percentage, proline, carbohydrates, gene expression, and activities of antioxidant enzymes. Moreover, a significant positive correlation was indicated between shoot dry materials with relative water content percentage, sucrose, catalase activity, ascorbate peroxidase activity, and cytosolic ascorbate peroxidase relative gene expression ratio. Antioxidant enzymatic activity and the association between gene transcript levels and activity seem to play roles in protecting plants from oxidative damage in drought stress. The lowest ascorbate peroxidase activity was measured under normal conditions in the cultivar Option (409 nmol ascorbate oxidation per min/g protein), and the highest ascorbate peroxidase was observed in RGS003 (600 nmol ascorbate oxidation per min/g protein) in high stress. Additionally, in the case of relative gene expression ratio the lowest cytosolic ascorbate peroxidase was observed under normal conditions in Option (1.05), and the highest cytosolic ascorbate peroxidase was seen in RGS003 (1.48) in high stress. According to the results of the current study, RGS003 seems to be more tolerant than Option.
Silymarin is one of the most potent antioxidant so far developed from plant sources used as hepatoprotectants. Influence of different concentrations (0, 1, 2, 4, 6 and 8mg/50ml culture) and exposure time (24, 48, 72, 96 and 120h) of salicylic acid on lipoxygenase activity, linoleic acid content, growth and production of silymarin in hairy root cultures of S. marianum were investigated. Detection and identification of flavonolignans was carried out by high performance liquid chromatograph method. Salicylic acid enhanced silymarin production (1.89mgg(-1) DW). The optimal feeding condition was the addition of salicylic acid (6 mg/50 ml culture) after 24h in which the silymarin content was 2.42 times higher than the control (0.78mgg(-1) DW). The content of silybin, isosilybin, silychristin, silydianin and taxifolin were 0.703, 0.017, 0.289, 0.02 and 0.863mgg(-1) DW respectively in these samples, while in non-treated hairy roots were 0.027, 0.046, 0.23, 0.022 and 0.453 respectively. Lipoxygenase activity also affected by elicitation. lipoxygenase activity increased 24h after treatment by approximately 1.57- fold (0.21 Delta OD(234)/mgproteinmin(-1)). Upon elicitation with salicylic acid, linoleic acid content of hairy roots (38.26mgg(-1) DW) were also elevated after 24h, in which the linoleic acid content was 2.37 times higher than the control (16.1mgg(-1) DW). It is feasible that elicitation with salicylic acid regulates the jasmonate pathway, which in turn mediates the elicitor-induced accumulation of silymarin.
