Allograft bone has been widely used for reconstruction of different portions of the skeleton. The fragment of bone harvested must be kept under low temperatures. The cryopreservation also contributes to decrease the antigenic potential of the tissue. Although this technique is considered safe, there is little information about the morphological modifications that the medullary and cortical portions of bone suffer after freezing. Hence, the aim of this study was to investigate the morphology of bone tissue after freezing under different temperatures and periods. Twelve rabbits were used to analyze the effects of two temperatures, -20 degrees C and -70 degrees C, during four periods of time: 30, 60, 90, 120 days. Tissues were analyzed by HE, picro-sirius stains and also by Feulgen's reaction, through qualitative and morphometric ways, considering the area occupied by cells and nuclei, medullary and cortical portions, as well as by collagen expression at cortical. The differences among the treatments were analyzed by Tukey s test, at 5% significance level. Bone freezing increased cellular and nuclear areas at cancellous bone and diminished nuclear area at the cortical bone. Cortical bone collagen suffered denaturation proportionally to temperature decrease and to freezing duration. These alterations compromised the morphology of tissues after 90 or 120 days of freezing at the temperature of -70 degrees C. Cells necrosed during freezing, contributing to reduce bone antigenicity.
Records of birth weight (BW) and weights standardized to 210 (W210); 365 (W365) and 550 (W550) of age in the Nellore breed were used to estimate variance components of genetic effects and predict genetic values of sires. The model included the fixed effects of contemporary group and cow age at calving as co-variable, and the additive and maternal genetic, permanent maternal environmental, and temporary environmental random effects. Estimates of means and standard deviations for BW, W210; W365 and W550 were 32.76 ± 3.74; 184.30 ± 29.02; 240.31 ± 41.85 and 322.12 ± 60.77, respectively. A significant additive genetic variability was detected for all weights. The permanent maternal environmental variance showed greater relevance on weaning weight, and it was practically inexistent after weaning. For the maternal genetic variance, the estimate for birth weight was more significant as compared with weaning weight. The estimated heritability values for BW, W210; W365 and W550 were 0.37 ± 0.02; 0.36 ± 0.03; 031 ± 0.01 and 0.38 ± 0.02, respectively. Genetic correlations between birth weight and other weights were of low magnitude, with high values of weights at other ages. The use of sires of higher genetic value for total maternal effect makes it possible to use sires that are more positive for weaning weight and also for year and yearling weight.
RESUMOForam desenvolvidos objetivos de seleção para gado de corte criado nas condições de produção típicas do Brasil Central. As características consideradas foram: número de bezerros desmamados por vaca/ano (NBD), peso da carcaça (PC), peso à desmama (PD) e consumo de alimento. Foram adotados dois sistemas de produção, um que considerava somente a cria e outro, o ciclo completo (cria, recria e engorda). No primeiro, as receitas foram a venda de bezerros à desmama, novilhas excedentes e vacas de descarte. No segundo, a receita foi a venda das novilhas excedentes, vacas de descarte e novilhos para o abate. Em ambas as situações, o lucro (USD/vaca/ano) foi estimado pela diferença entre receita e despesa. Os valores econômicos estimados foram expressos em dólar por unidade de mudança na característica, calculados na base vaca/ano. O valor econômico foi calculado avaliando-se a alteração ocorrida no lucro quando a característica era incrementada de uma unidade, permanecendo as demais inalteradas. A característica que apresentou maior valor bruto do ponderador econômico foi o NBD, seguida pelo consumo, PC e PD. Quando se considerou o valor econômico, em unidades de desvio-padrão genético-aditivo, a ordem de importância foi consumo, PC, NBD e PD.Palavras-chave: bovino de corte, objetivo de seleção, ponderador econômico ABSTRACT Breeding objectives were developed for beef cattle production under typical economic and environmental conditions of central Brazil. The traits considered were: calves weaned per cow/year (NCW), weaning weight (WW), carcass weight (CW) and food intake. In the study, two systems were examined, a cowcalf system (surplus calves sold after weaning) and a cow-calf enterprise (in which surplus calves are raised for slaughter). In the first, income is from the sale of male calves and surplus heifers after weaning and of culled cows. In the second, the income is from the sale of steers, surplus heifers and culled cows. For both situations, the profit (US$/cow/year) was estimated by the difference between revenue and costs. The economical values (EV) were calculated as the change in profit resulting from a unit change in each trait, as other traits remained unchanged. The trait with greatest economic value was NCW, followed by food intake, CW and WW. The order of importance when values were in additive genetic standard deviation units were feed intake, CW, NCW and WW.
