This experiment was conducted to study the effects of arginine supplementation on organ development, egg quality, blood parameters, and immune status of laying hens. A total of 360 25-week-old brown Leghorn laying hens were randomly divided into three groups with six replicates of 20 birds each and fed diets supplemented with 0, 8.5, or 17 mg of L-arginine/kg for 42 days. Results showed that the weight of proventriculus and duodenum in the treatment supplemented with 17 mg/kg L-arginine was heavier than that of 0 mg L-arginine/kg treatment (p<0.05). The weight of oviduct in the treatments supplemented with 17 mg/kg L-arginine was smaller than that of 0 and 8.5 mg L-arginine/ kg treatment (p<0.05), and the small yellow follicle amount in the treatment supplemented with 17 mg/kg L-arginine was less than that of 0 mg L-arginine/kg treatment (p<0.05), while its yolk color was deeper than that of 0 mg L-arginine/kg treatment (p<0.05), and the IgY content showed the same tendency. Total cholesterol and triglyceride levels in the treatment supplemented with 8.5 mg/kg L-arginine were lower than that of 0 mg L-arginine/kg treatment (p<0.05). The concentration of IL-2 in the treatment supplemented with 17 mg/ kg L-arginine were more than that of 0 mg L-arginine/kg treatment (p<0.05). The findings of this study show that 17 mg/kg L-arginine supplementation has beneficial effects onlayers' immune status and yolk IgY content, as well as on proventriculus and duodenum weight while no adverse effects were observed on laying performance, egg quality, or blood parameters.
This study was conducted to reconstruct heterogeneous embryos using equine skin fibroblast cells as donor karyoplasts and the bovine oocytes as recipient cytoplast for investigating the reprogramming of equine somatic cell nuclear in bovine oocyte cytoplasm and the developmental potential of the reconstructed embryos. Adult horse skin fibroblast cells serum-starved were used as donor somatic cells. Bovine oocytes matured in vitro were employed as recipient cytoplasts. The fusion of fibroblast cells into recipient cytoplasm was induced by electofusion. The fused eggs were activated by inomycin with 2 mm/ml 6-dimethylaminopurine (6-DMAP). The activated reconstructed embryos were co-cultured with bovine cumulus cells in synthetic oviduct fluid supplemented with amino acid (SOFaa) and 10% fetal calf serum (FCS) for 168 h. The results showed that the first completed cleavage of xenonuclear transfer equine embryos occurred between 30 and 48 h following activation. 52% of the injected oocytes were successfully fused, 72% of the fused eggs underwent the first egg cleavage and 17% of the heterospecific nuclear-transferred zygotes developed to 4- or 8-cell embryo stages. This study demonstrated that the reconstructed embryos have undergone the first embryonic division and the reprogramming of equine fibroblast nuclei can be initiated in bovine-enucleated oocytes.
A good understanding of water table fluctuation effects on vegetation is crucial for sustaining fragile hydrology and ecology of semiarid areas such as the Horqin Sandy Land (HSL) in northern China, but such understanding is not well documented in literature. The objectives of this study were to examine spatio-temporal variations of water table and their effects on vegetation in a semiarid environment. A 9.71 km<sup>2</sup> area within the HSL was chosen and well-instrumented to continuously measure hydrometeorologic parameters (e.g., water table). The area comprises of meadow lands and sandy dunes as well as transitional zones in between. In addition to those measured data, this study also used Landsat TM and MODIS imageries and meteorological data at a station near the study area. The spatio-temporal variations were examined using visual plots and contour maps, while the effects on vegetation were determined by overlaying a water table depth map with a vegetation index map derived from the MODIS imageries. The results indicated that water table was mainly dependent on local topography, localized geological settings, and human activities (e.g., reclamation). At annual and monthly scales, water table was mainly a function of precipitation and potential evapotranspiration. A region within the study area where depth to water table was smaller tended to have better (i.e., more dense and productive) vegetation cover. Further, the results revealed that water table fluctuation was more sensitive for vegetations in the meadow lands than in the transitional zones, but it was least sensitive for vegetations in the sandy dunes
