The B-cell receptor signaling pathway, which is critical to the development and maturation of normal B-cells, is emerging as an attractive therapeutic target in B-cell malignancies. Ibrutinib is a potent irreversible inhibitor of Bruton's tyrosine kinase (Btk), a key kinase important for signal transduction in the B-cell receptor (BCR) pathway. In preclinical studies, ibrutinib potently bound to Btk, inhibited BCR signaling, and decreased tumor cell proliferation and survival in many B-cell malignancy models. Excellent safety and efficacy data in clinical trials have led to US Food and Drug Administration (FDA) approval of ibrutinib for previously treated mantle cell lymphoma (MCL) and chronic lymphocytic leukemia (CLL), as well as CLL with 17p deletion. Ongoing clinical studies have also demonstrated great potency of ibrutinib in treating other types of non-Hodgkin's lymphoma (NHL), including diffuse large B-cell lymphoma (DLBCL), follicular lymphoma (FL), and Waldenström's macroglobulinemia (WM). Combination of ibrutinib with chemoimmunotherapy and other promising novel agents in B-cell malignancy therapy has also been under clinical investigation.
Daily treatment of postmenopausal Chinese women with DRSP/E2 for 16 weeks significantly reduced the incidence of hot flushes and demonstrated advantages vs. placebo for other climacteric symptoms. These results indicate that DRSP/E2 is effective, safe and well tolerated in postmenopausal Chinese women.
Light can have acute effects on human performance, including task performance, alertness and circadian phase shift. Most studies have investigated these effects using static light. This study investigates the effects of dynamic light with different cycle times and different ranges of correlated colour temperature on human alertness and task performance. Ten participants took part in the experiment using six conditions of dynamic light with each observing session lasting 4.5 hours. An electroencephelogram, measurements of critical flicker frequency, performance on various cognitive tasks and alertness and sleepiness questionnaires were used to evaluate the human responses. The results showed that participants appeared more alert and performed better under lighting of higher correlated colour temperature range but different correlated colour temperature cycle times had little effect.
The first two authors contributed equally to this study This study explores the inducing-apoptotic activity of the ethanol extract of Duchesnea indica Focke on treatment of herpes simplex encephalitis. Cell models were employed and divided into 4 groups: normal group, virus group, Duchesnea indica group and dexamethasone group. Cytopathic effect examination was employed to detect apoptosis of PC-t2 and BV-2 cells. ELISA was used to measure TNF-a and IL-t!J, and Greiss method to measure NO secretion. Flow cytometry assay for caspase-3 expressions was performed. As a result, the ethanol extract of Duchesnea indica could protect the neuron cell model from impairment by virus. In the cell model of microglia stimulated by herpes simplex virus (HSV), with the ethanol extract intervention, TNF-a, IL-t!J and NO levels were significantly decreased and cell death of BV-2 cells were markedly increased. The expression level of caspase-3 was notably elevated after the extract intervention. In conclusion, the ethanol extract of Duchesnea indica can reduce HSV-induced inflammatory injury on neuron due to the induction of microglia apoptosis.• Herpes simplex encephalitis (HSE), a kind of viral encephalitis, is a significant cause of morbidity and mortality, and often leaves patients long-term neurologic sequelae (I). Ingestion of one solution was paired with nutritive intragastric injections and ingestion of the other was paired with saline injections.
