BackgroundXiao-Yao-San (XYS) is a Chinese medicinal formula for treating anxiety and depression. This study aims to evaluate the use of a room-temperature super-extraction system (RTSES) to extract the major active components of XYS and enhance their psycho-pharmacological effects.MethodsThe neuroprotective roles of XYS/RTSES against reserpine-derived neurotoxicity were evaluated using a glial cell injury system (in vitro) and a depression-like C57BL/6 J mouse model (in vivo). The anxiolytic-behavioural effects were measured by the elevated plus-maze (EPM) test and the antidepressant effects were evaluated by the forced swimming test (FST) and tail suspension test (TST). Glucose tolerance and insulin resistance were assayed by ELISA. The expression of 5-HT1A receptors in the prefrontal cortex was examined by western blotting.ResultsXYS/RTSES (300 μg/mL) diminished reserpine-induced glial cell death more effectively than either XYS (300 μg/mL) or fluoxetine (30 μM) at 24 h (P = 0.0481 and P = 0.054, respectively). Oral administration of XYS/RTSES (500 mg/kg/day) for 4 consecutive weeks significantly elevated the ratios of entries (open arms/closed arms; P = 0.0177) and shuttle activity (P = 0.00149) on the EPM test, and reduced the immobility time by 90% on the TST (P = 0.00000538) and FST (P = 0.0000053839). XYS/RTSES also improved the regulation of blood glucose (P = 0.0305) and increased the insulin sensitivity (P = 0.0093). The Western blot results indicated that the activation of cerebral 5-HT1A receptors may be involved in the mechanisms of XYS/RTSES actions.ConclusionThe RTSES could provide a novel method for extracting effective anxiolytic- and antidepressant-like substances. XYS/RTSES improved the regulation of blood glucose and increased the insulin sensitivity in reserpine-induced anxiety and depression. Neuroprotection of glial cells and activation of cerebral 5-HT1A receptors were also involved.
Numerous materials have been proposed for bone tissue engineering. In this study, a newly designed hybrid composite scaffold composed of poly (D,L-lactide-co-glycolide) and a naturally bioceramic hybrid material, nanonized pearl powder, were prepared and the biological activities and physical properties of the scaffold for bone tissue engineering were evaluated. It is a composite consisting calcium carbonate crystal in an aragonite structure, embedded in an organic matrix. Peral contains one or more signal molecules capable of stimulating bone formation. The nanonized pearl powder is considered as a promising osteoinductive biomaterial. This biomaterial is biocompatible and shows osteogenic activity. In this study, the designed biohybrid of nanonized pearl powder/poly (lactide-co-glycolide) (NPP/PLGA) biocomposite scaffolds would employ biodegradable material as MC3T3-E1 cells seeded scaffolds. Therefore, the biocomposite scaffolds would be used to culture with MC3T3-E1 cells under spinner bioreactor in vitro. Furthermore, it also detailed how these tissues were characterized, qualitatively and quantitatively, with scanning electron microscopy and biochemical testing. The identity and the mode of action of these molecules on the osteoblast differentiation were analyzed. This study indicates that the efficiency of nanonized pearl powders in bone cell differentiation are certainly different from that of proteins. Further sudy will look forward to manufacturing the promising new generation bone substitute, three dimensional biocomposite scaffolds to replace the implant and autogeneous bone graft, which combines basic research and clinical application.
Many severe epidemics are caused by enteroviruses (EVs) and coronaviruses (CoVs), including feline coronavirus (FCoV) in cats, epidemic diarrhea disease virus (PEDV) in pigs, infectious bronchitis virus (IBV) in chickens, and EV71 in human. Vaccines and antiviral drugs are used to prevent and treat the infection of EVs and CoVs, but the effectiveness is affected due to rapidly changing RNA viruses. Many plant extracts have been proven to have antiviral properties despite the continuous mutations of viruses. Napier grass (Pennisetum purpureum) has high phenolic content and has been used as healthy food materials, livestock feed, biofuels, and more. This study tested the antiviral properties of P. purpureum extract against FCoV, PEDV, IBV, and EV71 by in vitro cytotoxicity assay, TCID50 virus infection assay, and chicken embryo infection assay. The findings showed that P. purpureum extract has the potential of being disinfectant to limit the spread of CoVs and EVs because the extract can inhibit the infection of EV71, FCoV, and PEDV in cells, and significantly reduce the severity of symptoms caused by IBV in chicken embryos.
