The aims of study was to compare the glycerol concentration in Tris glucose egg yolk (TGEY) diluents on the quality of deer frozen semen. Semen was collected from 5 Timor deer using electroejaculator. Immediately after collection the semen was evaluated macroscopic and microscopically. After initial evaluation, the semen was divided into three tubes and extended with Tris egg yolk with three different glycerol concentrations, which were 10% (TGEY 10); 12% (TGEY 12) and 14% (TGEY 14). The sperm motility, viability, acrosome intact and membrane intact were evaluated in raw semen, after equilibration and after thawing. The results showed that there were no differences (p>0.05) on the sperm motility, viability as well as sperm acrosome intact. Sperm membrane intact in TGEY 10 (52.50±5.89%) and TGEY 14 (51.50±4.12 %) were higher (p<0.05) than in TGEY 12 (49.00±6.58). It was concluded that 10, 12 or 14% glycerol concentration can be used for Timor deer semen cryopreservation.
ABSTRAKPenelitian ini bertujuan untuk menentukan waktu optimum untuk pengujian membran plasma yang utuh (MPU) pada sperma domba garut setelah inkubasi pada larutan hypo-osmotic. Sebanyak 24 ejakulat dari 8 ekor semen domba garut yang telah dewasa kelamin dikoleksi menggunakan vagina buatan. Semen dievaluasi secara makroskopis selanjutnya dilakukan pengujian MPU dengan mengambil 10 µL semen segar dan dimasukkan ke dalam 2 mL larutan hypo-osmotic kemudian diinkubasi pada suhu 37 o C selama 1 jam. Sample dievaluasi setiap 15 menit dengan menghitung 200 sperma yang bereaksi (coil) dan yang tidak bereaksi dari lima lapang pandang. Hasil penelitian menunjukkan waktu optimal pengujian MPU menggunakan larutan hypo-osmotic adalah 30 menit dan tidak ada perbedaan antar 8 domba jantan yang digunakan Kata Kunci : Keutuhan membran plasma, Semen domba garut, HOS test ABSTRACTThe study was aimed to determine the optimal time to evaluate the maximum numbers of swollen sperm after expose to hypo-osmotic solution for testing membrane integrity in fresh garut ram semen. A total of 24 ejaculate samples from eight sexually mature garut ram were collected using artificial vagina. After macroscopic evaluation, 10 µL undiluted semen was gently mixed in each of the 2 mL hypoosmotic solution and incubated at 37 o C for 1 hour. Two hundred of coil and non-coiled sperm were observed from five different fields every 15 minutes. Results demonstrated that the maximum percentage of coiling sperm positive to HOS test was at 30 minutes and no differences among 8 garut rams used.
ABSTRAK Penelitian ini dilakukan untuk mengetahui efektifitas pengencer Zorlesco dan modifikasi Zorlesco dengan beberapa jenis susu kacang kedelai dalam mempertahankan fertilitas spermatozoa babi landrace yang disimpan pada suhu 18⁰C. Semen dikoleksi dua kali seminggu menggunakan metode masase dari dua ekor pejantan landrace berumur ±2,5 tahun yang memiliki kodisi tubuh dan organ reproduksi yang sehat. Semen yang memenuhi syarat setelah di evaluasi secara makroskopis dan mikroskopis diencerkan dengan Zorlesco (P0) atau modifikasi Zorlesco dengan suplementasi susu kacang kedelai murni (P1), Metabolis (P2) dan Melilea (P3), dan masing-masing perlakuan diulang lima kali. Semen selanjutnya disimpan pada suhu 18⁰C. Data tentang motilitas dan viabilitas spermatozoa di analisis dengan ANOVA dan dilanjutkan dengan uji Duncan. Hasil analisis statistik menunjukkan bahwa modifikasi pengencer Zorlesco yang ditambahkan dengan susu kedelai 5% tidak dapat (P<0.05) mempertahankan motilitas dan viabilitas spermatozoa lebih tinggi dari pengencer Zorlesco selama 42 jam penyimpanan. Kata Kunci: susu kacang kedelai, modifikasi Zorlesco, motilitas Spermatozoa, viabilitas spermatozoa, babi landrace Sperm Fertility of Landrace Boar in Modified Zorlesco Extender with Soymilk ABSTRACT A research has been done to study the effectiveness of Zorlesco extender and modified Zorlesco extender with soymilk to maintain sperm fertility of landrace boar preserved at 18˚C. Semen was collected twice a week by hand massage from 2 heads of ± 2.5 years old landrace boar. A Completely Randomized Design with 4 treatments and 5 replications was used to organized the experiment. The fresh evaluated semen were extended with zorlesco P(0), zorlesco + hand made soymilk P(1), zorlesco + Metabolis TM soymilk P(2), and zorlesco + Melilea TM soymilk P(3). Extended semen were preserved at 18 0 C. Analysis of variance and Duncan test were used to analyzed data of motility and viability of sperm. The result showed that the modification of diluent Zorlesco added with 5% soymilk could not defend by sperm motility and viability spermatozoa compared with Zorlesco for 42 hours storage (P<0.05).
This study aimed to determine the effect of palmyra juice and sugarcane juice supplementation in durasperm diluent on the quality of Duroc boar liquid semen. The source of semen a from 3 Duroc boar aged 2-3 years, which were collected twice a week using the glove hand method. Semen was evaluated macroscopically and microscopically, good quality semen was divided into three parts and subjected to treatment: durasperm (PO), P1 durasperm + palmyra juice (PJ), P2 durasperm + sugarcane juice (SJ) and then stored in a Styrofoam box at 18 o -20 o C. Observations were made every eight hours. The data obtained were analyzed using Analysis of Variance (ANOVA) using the SPSS 20 program. The results showed that the percentage of motility at 64 hours showed that P1 was 40% higher (P <0.05) than P2 (35.00 ± 1.82) and P0 (27.50 ± 2.88). P1 had a higher percentage of spermatozoa viability 50.94 ± 0.79 (P <0.05) than P2 (45.50 ± 1.94) and P0 (37.75 ± 3.48). P1 has a higher MPU of 52.92 ± 0.61 with P2 (47.38 ± 1.47) and P0 (40.49 ± 3.47). Abnormality and pH were not significantly different (P˃0.05). It was concluded that palmyra juice supplementation diluent (P1) was effective in maintaining motility, viability, abnormalities, MPU and maintaining a stable pH.
A study was carried out in one of Indonesia native cattle, Sumba Ongole (SO) Cattle for determination of statuses of various biochemical parameters, aspartate aminotransferase (AST or SGOT) and alanine aminotransferase (ALT or SGPT). Apparently blood from randomly selected cattle (n= 36) were collected aseptically. Biochemical analysis of AST and ALT was performed on UPT. Laboratorium Kesehatan, Kupang, NTT. The results showed that the variations of parameters were not significant (p>0.05) between male and female SO cattle. The AST/SGOT (142.56±20.55 U/l) was slightly higher in female compared with the male (137.00±16.24 U/l), while level of ALT/SGPT were 46.44±10.99 U/l in male and 46.41 ± 10.23 U/l in female. Although, the result was not significant (p>0.05), level of AST and ALT in SO cattle was higher compared to standard which is 60-125 U/l for AST and 6.9-35 U/l for ALT. Results showed possibility of seasonal effect on AST/ SGOT and ALT/SGPT level in male and female SO cattle.
