As the precursor of vitamin A, β-carotene has a positive effect on reproductive performance. Our previous study has shown that β-carotene can increase antioxidant enzyme activity potentially through regulating gut microbiota in pregnant sows. This study aimed to clarify the effect of β-carotene on reproductive performance and postpartum uterine recovery from the aspect of inflammation and gut microbiota by using a mouse model. Twenty-seven 6 weeks old female Kunming mice were randomly assigned into 3 groups (n=9), and fed with a diet containing 0, 30 or 90 mg/kg β-carotene, respectively. The results showed that dietary supplementation of β-carotene reduced postpartum uterine hyperemia and uterine mass index (P<0.05), improved intestinal villus height and villus height to crypt depth ratio, decreased serum TNF-α and IL-4 concentration (P<0.05), while no differences were observed in litter size and litter weight among three treatments. Characterization of gut microbiota revealed that β-carotene up-regulated the relative abundance of genera Akkermansia, Candidatus Stoquefichus and Faecalibaculum, but down-regulated the relative abundance of Alloprevotella and Helicobacter. Correlation analysis revealed that Akkermansia was negatively correlated with the IL-4 concentration, while Candidatus Stoquefichus and Faecalibaculum had a negative linear correlation with both TNF-α and IL-4 concentration. On the other hand, Alloprevotella was positively correlated with the TNF-α, and Helicobacter had a positive correlation with both TNF-α and IL-4 concentration. These data demonstrated that dietary supplementation of β-carotene contributes to postpartum uterine recovery by decreasing postpartum uterine hemorrhage and inhibiting the production of inflammatory cytokines potentially through modulating gut microbiota.
Site-selective acetylation of a single lysine residue in a protein that reaches a lysine acetyltransferase’s accuracy, precision, and reliability is challenging. Here, we report a peptide-guided, proximity-driven group transfer reaction that acetylates a single lysine residue, Lys 248, of the fragment crystallizable region (Fc region) in the heavy chain of the human Immunoglobulin G (IgG). An Fc-interacting peptide bound with the Fc domain and positioned a phenolic ester close to Lys 248, which induced a nucleophilic reaction and resulted in the transfer of an acetyl group to Lys 248. The acetylation reaction proceeded to a decent yield under the physiological condition without the need for deglycosylation, unnatural amino acids, or catalysts. Along with acetylation, functional moieties such as azide, alkyne, fluorescent molecules, or biotin could also be site-selectively installed on Lys 248, allowing IgG’s further derivatization. We then synthesized an antibody–lipid conjugate and constructed antibody-conjugated liposomes (immunoliposomes), targeting HER2-positive (HER2+) cancer cells. We also built a bispecific antibody complex (bsAbC) covalently linking an anti-HER2 antibody and an anti-CD3 antibody. The bsAbC showed in vitro effector-cell-mediated cytotoxicity at nanomolar concentrations. Compared with bispecific antibodies (bsAbs), bsAbCs are constructed based on native IgGs and contain two antigen-binding sites to each antigen, twice that of bsAbs. Altogether, this work reports a method of site-selective acetylation of native antibodies, highlights a facile way of site-selective IgG functionalization, and underscores the potential of bsAbCs in cancer immunotherapy.
In this work, we demonstrated a strategy to design a modified starch/polyvinyl alcohol composite (CCSP), which was employed as a highly efficient and economical fixed-bed adsorbent for treating textile wastewater. Characterization revealed that most of the CCSP was shaped with the morphology of sphericity, and had some water swelling properties. The crystallinity of the CCSP was lower than that of native starch and polyvinyl alcohol, and its average particle size gradually increased with the dosage increase of cationic starch in the preparation. Adsorption experiments showed that the adsorption capacities of CCSP were more than 605 and 539 mg/g for Reactive Black 5 and Reactive Orange 131, respectively, which were over 10 times larger than that of commercial activated carbon (AC). The mixture adsorbent composed of CCSP and AC could remove starch, polyvinyl alcohol, and dyes from textile wastewater completely and simultaneously combined with the fixed-bed technique, and its adsorption capacity was conducted as a function of the bed height and flow rate. Most importantly, the disabled mixture adsorbent could be converted into regenerated AC through a chemical activation process, thereby avoiding the production of solid waste. This study will provide a new efficient green sustainable method for treating textile wastewater.
The energy and metabolic state of sows will alter considerably over different phases of gestation. Maternal metabolism increases dramatically, particularly in late pregnancy. This is accompanied by the development of an increase in oxidative stress, which has a considerable negative effect on the maternal and the placenta. As the only link between the maternal and the fetus, the placenta is critical for the maternal to deliver nutrients to the fetus and for the fetus’ survival and development. This review aimed to clarify the changes in energy and metabolism in sows during different pregnancy periods, as well as the impact of maternal oxidative stress on the placenta, which affects the fetus’ survival and development.
Objective: To explore the relationship between symptomatic functional status and quality of life of patients with low back myofascial pain syndrome (MPS). Methods: From July 2021 to June 2022, 106 patients with low back myofascial pain syndrome in the Affiliated Hospital of Hebei University were selected as the research subjects. A total of 106 MPS patients were investigated with general information questionnaire, Memory Symptom Assessment Scale (MSAS), Oswestry Disability Index (ODI) and Short Form Questionnaire (SF-36). The relationship between quality of life and symptom distress and dysfunction was observed and analyzed based on symptom distress and dysfunction scores, SF-36 scores, and so on. Results: The total score of MSAS was 1.79 ± 0.91. The overall symptom distress of the patients was moderate. The ODI score was 18.46 ± 5.95. The functional disability of the patients was classified as moderately impaired. The MSAS-PHYS, MSAS-PSYCH, MSAS-GDI three scale scores were 2.14 ± 0.75, 1.69 ± 0.88, 1.55 ± 0.46, respectively, and the variability of the three scales is relatively large; the dimension scores were significantly lower than those of the conventional scoring models, and P < 0.05, indicating a statistical difference; the scores of each dimension of the patient’s quality of life were compared with the scores of symptom distress and functional status. The higher the symptom distress score, the lower the quality of life, with P < 0.05, indicating a statistical difference; the higher the score of each dimension of functional status, the better the quality of life, showing a positive correlation, and P < 0.05, indicating a statistical difference. Conclusion: MPS patients face a number of physical and psychological symptoms, and their functional status is limited. Nursing staff should implement health education and intervention measures according to the actual situation of the patients, so as to improve the quality of their lives.
