The COVID-19 pandemic caused by the SARS-CoV-2 virus continually poses serious threats to global public health. The main protease (Mpro) of SARS-CoV-2 plays a central role in viral replication. We designed and synthesized 32 new bicycloproline-containing Mpro inhibitors derived from either Boceprevir or Telaprevir, both of which are approved antivirals. All compounds inhibited SARS-CoV-2 Mpro activity in vitro with IC50 values ranging from 7.6 to 748.5 nM. The co-crystal structure of Mpro in complex with MI-23, one of the most potent compounds, revealed its interaction mode. Two compounds (MI-09 and MI-30) showed excellent antiviral activity in cell-based assays. In a SARS-CoV-2 infection transgenic mouse model, oral or intraperitoneal treatment with MI-09 or MI-30 significantly reduced lung viral loads and lung lesions. Both also displayed good pharmacokinetic properties and safety in rats.
Efficient organic emitters in the deep‐red region are rare due to the “energy gap law”. Herein, multiple boron (B)‐ and nitrogen (N)‐atoms embedded polycyclic heteroaromatics featuring hybridized π‐bonding/ non‐bonding molecular orbitals are constructed, providing a way to overcome the above luminescent boundary. The introduction of B‐phenyl‐B and N‐phenyl‐N structures enhances the electronic coupling of those para‐positioned atoms, forming restricted π‐bonds on the phenyl‐core for delocalized excited states and thus a narrow energy gap. The mutually ortho‐positioned B‐ and N‐atoms also induce a multi‐resonance effect on the peripheral skeleton for the non‐bonding orbitals, creating shallow potential energy surfaces to eliminate the high‐frequency vibrational quenching. The corresponding deep‐red emitters with peaks at 662 and 692 nm exhibit narrow full‐width at half‐maximums of 38 nm, high radiative decay rates of ca. 108 s−1, ≈100 % photo‐luminescence quantum yields and record‐high maximum external quantum efficiencies of ca. 28 % in a normal planar organic light‐emitting diode structure, simultaneously.
Background: Sarcopenia is a condition that is characterized by loss of muscle mass, muscle strength and muscle functional impairment with ageing. It is associated with poor health outcomes, premature death and a significant burden on the global health economy. The prevalence of sarcopenia in China is unknown since most of the studies are lack of uniform standard. The study was undertaken to study the prevalence of sarcopenia and the association with cognitive impairment among multi-ethnic adults aged 50 years old or older in western China. Methods: We measured gait speed, handgrip strength and muscle mass by using bioelectrical impedance analysis (BIA) for all eligible participants and 4500 participants were eligible for the analysis. We defined sarcopenia using the diagnostic algorithm recommended by the Asian Working Group for Sarcopenia (AWGS). We assessed the participants' cognitive functions using the 10-item Short Portable Mental Status Questionnaire (SPMSQ). Relationships between sarcopenia and cognitive impairment were analyzed using univariate and multivariate analyses. Results: Of 4500 participants (mean age 62.4 ± 8.3 years), 869 (19.31%) adults were sarcopenia. 446 (9.9%) participants were identified as having mild cognitive impairment, 144 (3.2%) adults were identified as having moderate/severe cognitive impairment. After adjusting for age, gender, ethnics and other potential cofounders, cognitive impairment was found to be independently associated with sarcopenia with a dosage effect (mild cognitive impairment: odds ratio [OR]: 1.41, 95% CI 1.10-1.82; moderate/severe cognitive impairment: OR: 3.05, 95% CI 2.08-4.49). After gender stratification, the association between mild cognitive impairment with sarcopenia in male is not significant, while is still significant in female. While the association between moderate/severe cognitive impairment is independently associated with sarcopenia in both male and female. Conclusions: The prevalence rates of sarcopenia, mild cognitive impairment, moderate/severe cognitive impairment among the communities aged 50 or older in western China were 19.31, 9.9 and 3.2%, respectively. Cognitive impairment was significantly associated with sarcopenia with a dosage effect, especially in female.
Fluorinated graphene (FG) can be regarded as the representative two-dimensional (2D) material to study the characteristics of "2D chemistry", whereas its derivative reaction mechanism is still required to be revealed for the destination of deciduous fluorine atoms after defluorination of FG. Herein, we proposed a particular derivative reaction of FG by employing 2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPO) as the attacking reagent, and the products were characterized via Electron Paramagnetic Resonance Spectroscopy (EPR), Mass Spectrometry (MS) and X-ray photoelectron spectroscopy (XPS). It was demonstrated that the defluorination caused by TEMPO occurred in a radical mechanism, thus leading to formations of new spin centers on graphene nanosheets as well as C[double bond, length as m-dash]C bonds. More importantly, the deciduous fluorine atoms after defluorination, which existed in TEMPO fluoride molecules, have been detected for the first time. Meanwhile, some TEMPO molecules were covalently grafted on the nanosheet, which resulted from the coupled reaction between TEMPO radical and the spin center on the FG nanosheet. These findings deepen the research of derivative reactions of FG, meanwhile providing a particular view to investigate the chemistry characteristics of 2D materials from a radical mechanism.
