The presented here technique is developed for in vitro estimation of the influence of external con ditions on the deformation ability of human red blood cells. The method is based on the use of single beam laser trap for capturing of erythrocytes fixed to the bottom of the liquid cell by the adhesion force. The approach is suitable for evaluation of the properties of individual cells as well as for real time acquisition of the data on ensemble of erythrocytes large enough for subsequent statistical analysis. It is shown that both a change in the saline solution concentration and presence of certain stabilizing agents lead to measurable shift of the maximum of the cells distribution by their deformability.
Abstract:The decrease in clinical efficacy of oxygen therapy takes place in patients suffering from chronic pulmonary obstructive disease who continue smoking even if the intensity of smoking is getting lower. It is explained by the damage of oxygen utilization regardless of normalization of arterial blood saturation indices and perfusion oxygen saturation indices in blood flow.
Introduction. Throughout history, humanity has lived and developed in accordance with its biological rhythms, which provided the adaptation of the body to the factors of external and internal environment, ensuring the normal synchronous functioning of all its components.The purpose of this work was to study the stressful influence of circadian rhythms disturbance on patients with metabolic syndrome (MS) under conditions of dark deprivation.Material and methods of investigation. We selected patients with MS and disturbed circadian rhythms – a total of 102 people (all men) from among those who constantly work at night, in whom a decrease in melatonin levels was recorded at the preliminary stage. According to the set objectives, 102 patients (young age according to WHO criteria) were divided into three groups of 33, 37 and 32 persons according to the criterion "duration of influence of dark deprivation and MS history duration": 0–5, 5–10 and over 10 years. In the first group the mean age was 30.1 ± 1.4 years, in the second group – 36.5 ± 1.5 years, in the third group the mean age did not exceed 40.3 ± 0.9 years. Melatonin levels (as confirmation of circadian rhythm disturbances) in blood serum were studied in all study participants using Melatonin ELISA immunoassay kit (IBL, Germany). Results and discussion. Normally, when blood cortisol is measured before 10 a.m., its average value varies in a wide range from138 to 635 nmol/l. The increase up to 700 nmol/l is admitted, which is regarded as a negative influence of external factors during the examination. However, one should keep in mind that during evening measurements, cortisol levels in men may normally decrease to 80 nmol/l.Conclusions. Thus, dark deprivation in patients with metabolic syndrome leads to the formation of a stress response. Maximum changes occur after 10 years of disturbance of regulatory mechanisms and lead to a pronounced imbalance between the sympathetic and parasympathetic nervous system, increased vascular tone and changes in the profile of blood pressure during the day towards the predominance of Non-dippers and Night-peers, which increases the risk of progression of metabolic syndrome symptoms.

Цель исследования -изучение влияния транскраниальной электростимуляции (ТЭС-терапии) на состояние микроциркуляторного кровотока у пациентов с соматической коморбидной патологией -хронической обструктивной болезнью легких и системной артериальной гипертензией. Материал и методы. Исследовалась степень нарушения процессов микроциркуляции с использованием лазерной флоуметрии на диагностическом комплексе «ЛАКК-М». Оценивались среднее значение перфузии, сатурация смешанной (капиллярной) крови, сатурация артериальной крови, относительный объем фракции эритроцитов в коже подушечек среднего пальца, индекс перфузионной сатурации кислорода в микрокровотоке, индекс удельного потребления кислорода в ткани, эффективность кислородного обмена, флуоресцентный показатель потребления кислорода. Для коррекции выявленных нарушений в течение 1,5 года применялась транскраниальная электростимуляция. Результаты и их обсуждение. Под воздействием лечения у пациентов с коморбидной патологией снижался индекс перфузионной сатурации в микрокровотоке в среднем на 9% (≤0,05), происходило увеличение потребления кислорода в тканях, повышалась эффективность его обмена, снижалась активность гликолитических процессов. Отмечалось повышение флуоресцентного показателя потребления кислорода на 6,2, 9,8 и 9,0% (р 1-3 ≤0,05) соответственно в зависимости от длительности анамнеза хронической обструктивной болезни легких -до 5, 10 или 15 лет. Выводы. Влияние транскраниальной электростимуляции на микроциркуляторные нарушения реализуется через увеличение длительности адаптационных механизмов, направленных на борьбу с гипоксией на фоне коморбидной патологии, особенно при длительности анамнеза заболеваний менее 10 лет. Увеличение емкости микроциркуляторного русла за счет снижения тонуса прекапилляров и открытия резервных микрососудов способствует снижению системного артериального давления на фоне коморбидной патологии.
