Abstract: Linum flavum hairy roots were initiated from leaf discs using Agrobacterium rhizogenes strains LBA9402 and TR105 though two other strains, 15834 and A4, were relatively ineffective for induction. Significant variation in coniferin accumulation was observed between hairy root lines originating from different L. flavum seedlings and/or A. rhizogenes strains. Coniferin reached 58 mg g-1 dry wt by culturing the roots in Linsmaier and Skoog (LS) medium with 2,4-dichlorophenoxyacetic acid and naphthaleneacetic acid … Show more

“…In Linum flavum , A . rhizogenes strains LBA 9402 and TR 105 induced hairy roots with 50% frequency from leaf discs whereas low induction was observed with strains 15834 and A4 [24] . Variable degree of transformation efficiency was also observed in Beta vulgaris transformed with different Agrobacterium strains [25] and from root segments of Maytenus senegalensis using strains LBA 9402 and A4T [26] .…”

“…HR phenotype is characterized by vigorous growth, lack of geotropism, high lateral branching and genetic stability in hormone free medium [ 30 ]. L. flavum HR lines have been previously established and described as an effective system for the production of ATL including up to 3.5% DW of MPT [ 25 , 31 ] or lignan precursor such as coniferin [ 32 ]. However, in these studies few independent HR lines have been generated and analyzed while in the absence of a thorough assessment of the influence of growing conditions on ATL accumulation it is difficult to get a clear idea of the production potential of this species.…”

“…[16] In L. flavum , hairy roots were initiated from leaf discs with a success rate of approximately 50% using A. rhizogenes strains ‘LBA9402’and ‘TR105’. [6] Evidently, the selection of an effective bacterial strain for the production of transformed root cultures is highly dependent on the plant species, and must be determined empirically. [17] The bacterial strains used in this study exhibit different levels of virulence to this plant species and the induction of hairy root occurred at different time depended on strains.…”
