A Method to Monitor the Introduction of Posttranscriptional Modifications in tRNAs with NMR Spectroscopy

Abstract: During their biosynthesis, transfer RNAs (tRNAs) are decorated all along their sequence with a large number of posttranscriptional chemical modifications. Methods to directly detect the introduction of posttranscriptional modifications during tRNA maturation are rare and do not easily bring information on the temporality of modification events. Here, we report a methodology, using NMR as a tool to monitor tRNA maturation in a nondisruptive and continuous fashion in cellular extracts. This method requires the p… Show more

“…For instance, in the simple case where m 1 A58 would be introduced before m 5 Cs, there could not be any observable effect of m 5 Cs on the introduction of m 1 A58. In order to investigate whether this positive effect remains in a cellular context, we undertook to apply our recently developed methodology (38,39) to the monitoring of m 1 A58 introduction in tRNA i Met in yeast extracts. As seen above, the imino signals of G18 and U55 constitute an NMR signature of a properly folded elbow structure, and therefore can be regarded as an indirect marker of m 1 A58 introduction in the case of tRNA i Met .…”

“…In addition, previous time-resolved NMR study of tRNA Phe in pus4Δ and trm2Δ yeast extracts pointed towards the m 1 A58 incorporation being more affected by Ѱ55 than by T54, which is perfectly in line with the kinetic data reported here. This indicates that the time-resolved NMR approach we have developed in cellular extracts (39), is not only reliable to identify cross-talks between modifications, but also to discriminate between weak and strong dependencies.…”
