A New Splice Variant of Glial Fibrillary Acidic Protein, GFAPε, Interacts with the Presenilin Proteins

Abstract: We describe a new human isoform, GFAP⑀, of the intermediary filament protein GFAP (glial fibrillary acidic protein). GFAP⑀ mRNA is the result of alternative splicing and a new polyadenylation signal, and thus GFAP⑀ has a new C-terminal protein sequence. This provides GFAP⑀ with the capacity for specific binding of presenilin proteins in yeast and in vitro. Our observations suggest a direct link between the presenilins and the cytoskeleton where GFAP⑀ is incorporated. Mutations in GFAP and presenilins are assoc… Show more

“…Based on the mass spectral analysis, it appears that only isoform 2 is present in the storage bodies. A number of GFAP splice variants have also been reported in humans and other species (Condorelli et al 1999;Nielsen et al 2002;Singh et al 2003). Canine isoform 2 corresponds to the isoform that is normally expressed predominantly in humans (Fuchs and Weber 1994;Reeves et al 1989).…”

“…For example, GFAPa is known to be the dominant isoform in the central nervous system. In Northern blot analysis of U373MG cells, two isoforms, known as GFAPa and e, had sizes of 2.9 kb and 1.8 kb, respectively (Nielsen et al, 2002). Levels of both mRNAs were decreased in U373MG-IE1 cells by approximately 15-to 20-fold, as determined by phosphoimage analysis (Fig.…”

“…The 2.9-kb astrocytic mRNA of GFAP encodes the dominating GFAP isoform (GFAPα) in the CNS [3,4]. Four other isoforms have been described, termed GFAPβ, γ, δ and ε, and at least ε is functionally distinct [7]. GFAP is a member of the class III intermediate filament proteins, which have a characteristic structure composed of a highly conserved central α-helical rod domain flanked by non-helical N-terminal head and C-terminal tail domains [8][9][10][11].…”
