A Newly Isolated Penicillium oxalicum 16 Cellulase with High Efficient Synergism and High Tolerance of Monosaccharide

Abstract: Compared to Trichoderma reesei RUT-C30 cellulase (Trcel), Penicillium oxalicum 16 cellulase (P16cel) from the fermentation supernatant produced a 2-fold higher glucose yield when degrading microcrystalline cellulose (MCC), possessed a 10-fold higher β-glucosidase (BGL) activity, but obtained somewhat lower other cellulase component activities. The optimal temperature and pH of β-1,4-endoglucanase, cellobiohydrolase, and filter paperase from P16cel were 50-60 °C and 4-5, respectively, but those of BGL reached 7… Show more

“…Endophytic fungi can combine with other endogenous microorganisms and antibacterial compounds to form a defense system that produces alkaloids to strengthen immunity and maintain growth under stress (Qin et al 2011; Clay and Holah 1999). Some endophytic fungi produce active proteases helped maintain plant activities, such as pectinase and esterase, which degrade cell walls (Zhao et al 2016). Pathology can result when programmed senescence in the plant or environmental change perturb the fungal population in the plant (Stamford et al 2001).…”

“…DNS spectrophotometry is a widely accepted and applied method in research (Aoyama et al, 2015;Miller, 1959;Wood and Bhat, 1988;Xiao et al, 2004;Zhao et al, 2016), which determined the ability of DNS to saccharify CMC via the analysis of its hydrolysis products, such as glucose and cellobiose (reducing sugars).…”

“…Cell-free medium was collected after centrifugation at 6000 g per 20 min. Enzyme activity was measured according to Zhao et al (2016) with some modifications. Endoglucanase activity was determined by incubating 900 μL of CMC (1%) with enzymatic extract to a final volume of 1.1 mL containing 0.1 M acetate buffer, pH 6.0, at 50 °C for 50 min.…”
