Vero cells have been derived from the kidney of African green monkeys (Cercopithecus aethiops) as mammalian continuous cell lines for biological research. Continuous culturing of Vero cells as well as maintenance of frozen stock is important for future use. Dimethyl sulphoxide (DMSO) and glycerol are two common cryoprotectants used widely to maintain frozen cell lines. DMSO, as a cryoprotectant in cryopreservation procedures, is detrimental to cells. A study was carried out to compare the effect of DMSO and glycerol as cryoprotectants on Vero cells. Vero cells were cultured and maintained using same nutrients and environment, following the modified protocol of Ammerman et al. (2008). After total cell counting the Vero cells were preserved in Cryocane at liquid nitrogen temperature (-196ºC). Two frozen stocks were made simultaneously from the cell suspensions of same concentrations using DMSO and glycerol. After one year of preservation post-thawed cell viability with glycerol was higher than with DMSO. Different methods for snap freezing surgical human tissue specimens exist. At pathology institutes with higher work loads, solid carbon dioxide, freezing sprays, and cryostat freezing are commonly used as coolants for diagnosing frozen tissue sections, whereas for tissue banking, liquid nitrogen or isopentane cooled with liquid nitrogen is preferred. Freezing tissues for diagnostic and research purposes are therefore often time consuming, laborious, even hazardous, and not user friendly. In tissue banks, frozen tissue samples are stored in cryovials, capsules, cryomolds, or cryocassettes. Tissues are additionally embedded using freezing media or wrapped in plastic bags or aluminum foils to prevent desiccation. The latter method aggravates enormously further tissue handling and processing. Here, we describe an isopentane-based workflow which concurrently facilitates tissue freezing and processing for both routine intra-operative frozen section and tissue banking and satisfies the qualitative demands of pathologists, cancer researchers, laboratory technicians, and tissue bankers.

Aim: The aim of the study was to investigate the histological changes of rat epididymis with the effect of cryopreservation on 15, 30, 45 and 60 days freezingthawing. Methods:The study was performed on male rats, epididymides collected were immediately subjected to 15, 30, 45 and 60 days freezing process, after completion of preservation time they were thawed at 37°C for 10' were fixed in formalin for histological investigation.Results: After15, 30, 45 and 60 days of cryopreservation rat epididymis showed reduce in number of sperm in lumen, degenerative changes in tissue shows elongated tubules and loss of epithelium and connective tissue. Conclusion:In conclusion, the histological changes obtained after freezing storage were reasonable; however, the cryopreserved tubules sheets could not maintain their original layered structure. This work needs to be done to better preservation of the rat epididymis.
