A novel near-infrared fluorescent probe with a “donor–π–acceptor” type structure and its application in the selective detection of cysteine in living cells

Abstract: A novel “donor–π–acceptor” type fluorescent probe has been prepared for the detection of Cys. This probe shows great fluorescent performance and obvious response to Cys, and has been successfully applied in visualizing Cys in living cells.

“…Miscellaneous Probes. Gong et al 124 have synthesized NIR-45, which is based on a donor-π-acceptor structure for the detection of Cys in PBS/DMF (1:1, v/v). A malononitrile unit was used as an acceptor, whereas a phenolic group (obtained after reaction with Cys) acts as a donor unit (Scheme 46).…”

“…[3][4][5][6] For example, a high concentration of Cys can cause neurocytotoxicity, while a low concentration of Cys can cause drowsiness, discoloration of hair, liver damage, etc. [7][8][9] The high concentration of Hcy in human blood is associated with cardiovascular disease and Alzheimer's disease. 10,11 The abnormal level of GSH is closely tied to the loss of white blood cells and cancer.…”

“…[30][31][32] Several NIR Cys probes have been reported recently. [33][34][35][36][37][38] For example, McCarley's group 39 reported dicyanomethylene-4Hpyrans (DCM dyes)-based with attractive photophysical properties NIR probe for detecting of Cys over Hcy, and GSH. Yang's group 40 has presented a ratiometric two-photon uorescent probe for detection Cys based on intramolecular charge transfer (ICT) mediated two-photon-FRET integration mechanism.…”
