A preparative isolation and purification of arctigenin and matairesinol fromForsythia koreana by centrifugal partition chromatography

Abstract: Centrifugal partition chromatography was applied to separate arctigenin and matairesinol from Forsythia koreana extract with a two-phase solvent system composed of n-hexane-ethyl acetate-methanol-water (5:5:5:5 v/v). Using this method, arctigenin and matairesinol were successfully separated from partially purified F. koreana extracts in only one step. The purities of isolated compounds were determined to be over 90% by HPLC analysis.

“…Arctigenin indicates a representative dibenzylbutyrolactone lignan with anti-tumor, anti-viral, and Ca 2+ antagonist activities (Kim et al, 2006), and a suppressive agents at the level of the activation of heat shock transcription factor, the induction of mRNA, and the synthesis and accumulation of heat shock proteins, which are involved in protecting cells and in the pathophysiology of diseases such as inflammation, cancer, and neurodegenerative disorders (Ishihara et al, 2006).…”

“…Many natural products have been purified successfully by CPC (Kim et al. , 2006; Chen and Wang, 2009). Previously, 5‐CQA has been purified by a number of different methods from many plants, such as Lonicera spp.…”

“…Arctigenin of 102 mg (with a purity of 98.9%) was obtained from 200 mg of hydrolyzed sample, and the recovery value was 89.6%. The purity of arctigenin by this method is higher than that in previous studies [18,19]. According to above process, 24.48 mg of arctigenin could be obtained from 1 g of powered Fructus Arctii.…”

“…The K D values were measured and are summarized in Table 1. A similar solvent system as that in Kim's study [19] (petroleum ether/ethyl acetate/methanol/water, 10:10:10:10, v/v) is not suitable for this separation. When petroleum ether/ethyl acetate/methanol/water at a ratio of 10:25:15:20 v/v was used as the two-phase solvent system, the K D values were suitable and arctigenin could be well separated from the other compounds.…”
