A Simple Competitive Protein-Binding Assay for Cyclic Adenosine 3´:5´-monophosphate

Abstract: A cyclic adenosine 3': 5'-monophosphate (Ado-3':5'-P) assay is proposed basing on the principle of competitive protein-binding. Free and bound Ado-3':5'-P is separatedjby coated (Dextran T 70, bovine serum albumine) charcoal. The Ado-3':5'-P binding protein is obtained from bull adrenals. The detection limit is 0.25 pmol in the incubation tube. The affinity constant of the protein-Ado-3': 5'-P complex is 2.8 • 10 8 l/mol. The free enthalpy is approximate lOkcal/mol. The high specificity is tested with other si… Show more

“…Cyclic AMP in unprocessed CSF was measured by a competitive protein-binding assay (Cooper et at., 1972) using a binding protein prepared as described by Wombacher and Koerber (1972). The cyclic AMP concen trations were expressed as picomoles cyclic AMP per milliliter.…”

“…The proteins were quantified by the method described by Lowry et al (1951). The levels of cyclic AMP in each sample were measured by the protein binding assay method described by Wombacher and Korber (1971) The maximum accumulation of cyclic AMP in the mature oocytes of grey mullet was recorded for total levels of 15 fL9 LH or SG-G100. This level was comparable to the pituitary contents reported in N. crysoleucas, which ranged between 6.1-34.0 fL g of SG-G100 during the 24-hr cycle (de Vlaming and Vodicnik, 1977).…”

“…Cyclic AMP in rat urine was measured by a competitive protein binding assay (COOPER et al 1972) using a binding protein prepared as described by WOMBACHER & KOERBER (1972). In human investigations urinary cyclic AMP was determined with a protein binding assay using a binding protein prepared from rabbit skeletal muscle essentially as described by MIYAMOTO et al (1969).…”
