A Vertebrate-Specific Chp-PAK-PIX Pathway Maintains E-Cadherin at Adherens Junctions during Zebrafish Epiboly

Abstract: BackgroundIn early vertebrate development, embryonic tissues modulate cell adhesiveness and acto-myosin contractility to correctly orchestrate the complex processes of gastrulation. E-cadherin (E-cadh) is the earliest expressed cadherin and is needed in the mesendodermal progenitors for efficient migration [1], [2]. Regulatory mechanisms involving directed E-cadh trafficking have been invoked downstream of Wnt11/5 signaling [3]. This non-canonical Wnt pathway regulates RhoA-ROK/DAAM1 to control the acto-myosin… Show more

“…PAK interacts with the Rac/Cdc42 GEF b-PIX, and depletion of b-PIX induced a similar gastrulation phenotype to RhoV depletion, suggesting that RhoV acts via PAK and b-PIX to localize E-cadherin. 39 So far RhoV has not been reported to affect E-cadherin localization in cultured cells in vitro, and thus this would be an interesting area for future research.…”

“…39 During gastrulation, the zebrafish embryo undergoes a series of morphological changes to form the 3 distinct germ layers: endoderm, mesoderm and ectoderm. E-cadherinmediated cell-cell adhesion is essential during this process.…”

“…Similarly, Rac1 and/or Cdc42, and their PAK effectors have been reported to bind to or affect GIT-PIX. But, in addition to PIX binding to inactive Rac1 and/or Cdc42 during activation, active Rac1 and/or Cdc42 (and other Rho family members) have been reported to also bind to GIT-PIX (Brown et al, 2002;Shin et al, 2004;Gu et al, 2006;ten Klooster et al, 2006;Hajdo-Milasinovic et al, 2009;Tay et al, 2010;Wilson et al, 2014). The effects binding of active Rho family proteins has to GIT-PIX remain illdefined and appear somewhat contradictory because GIT and PIX have each been reported to directly bind Rac1 and/or Cdc42 in cellbased studies where both are present, clouding interpretation.…”

“…Since NC cells form at the boundary of neural and nonneural ectoderm, it is possible that the regulation of cadherinmediated cell adhesion contributes to the process of NC fate specification, in addition to its known function in NC migration (Kashef et al, 2009;Park and Gumbiner, 2010;Taneyhill, 2008). Of note, both Wnt11 signaling and the RhoV/Chp GTPase, which is required for NC formation, have been implicated in the maintenance of adherens junctions in zebrafish embryos (Tay et al, 2010;Ulrich et al, 2005). Further studies are needed to examine the functions of cadherins and other cell junction components in NC specification.…”
