Diversity and morphological disparity of desmid assemblages in Central European peatlands

Morphological disparity has increasingly been used as an alternative measure of biological diversity based on the shape features of organisms. In this study, we investigated the species diversity and morphological disparity of benthic Desmidiales in Central European peatland pools. The shape features of cells were determined using the 3-D elliptical Fourier analysis of their frontal and lateral views. The resulting morphospace was used to calculate the contributions of localities and species to the morphological variation. In addition, the disparity of samples and their average cell complexity (indicating intricacy of cell shapes) was evaluated. These data were related to species diversity data and to the abiotic factors. Species diversity was positively correlated with pH and conductivity. The low-pH localities generally supported a more variable species composition than did slightly acidic to neutral localities.Conversely, the total nitrogen concentrations of these areas negatively correlated with species diversity. Interestingly, partial morphological disparity (measuring the contribution of a sample to the overall morphological variation) did not correlate with species diversity. On the contrary, several mountain peat bog localities had high disparity values, irrespective of their rather low species diversity. In addition, several samples from minerotrophic fens with high diversity had average or low values of partial morphological disparity. These results indicate the relative importance of mountain peat bogs for the total morphological diversity of Desmidiales within the region that could not be ascertained solely from species diversity data. The inner morphological disparity of samples was highly correlated with their species diversity. Species of the genus Micrasterias, Hyalotheca dissiliens and Desmidium species had the highest partial morphological disparity, thus indicating their marginal position within the morphospace. Micrasterias and Euastrum species had the highest complexity values. The average cell complexity of individual samples did not correlate with their diversity or disparity; however, it was positively correlated with the levels of total nitrogen and phosphorus, and illustrates a pattern different from that arrived at by species diversity data.

Biological invasions have attracted particular attention since they often result in serious consequences for natural ecosystems. One planktonic invasive species is Cylindrospermopsis raciborskii, a cyanobacterium originally reported to occur exclusively in the tropics. Over the last few decades its range has extended to temperate regions and it occupies shallow highly eutrophic lakes previously dominated by other cyanobacteria. The purpose of this study was to examine the ecology of C. raciborskii during Planktothrix agardhii blooms in two shallow lakes in western Poland and to determine whether these species have different environmental preferences. Multiple linear regression showed that the biomass of P. agardhii was significantly negatively related to Secchi depth in Lake Bnin´skie. In Lake Bytyn´skie, P. agardhii was significantly positively related to concentrations of PO 3À 4 , chlorophyll a and total phosphorus and negatively related to Secchi depth, NO À 3 , and total nitrogen. Cylindrospermopsis raciborskii was significantly positively related only to concentrations of NH þ 4 . There was a negative correlation between the biomass of P. agardhii and C. raciborskii perhaps showing different responses to environmental variables. Moreover, the biomass of P. agardhii was negatively correlated with Shannon-Wiener diversity of the phytoplankton assemblages. Our results support the concept that these cyanobacterial species have different environmental preferences and their niches differ from each other. These results suggest that light is an important driver of phytoplankton community structure resulting in shifts from a community dominated by P. agardhii in very turbid waters to more diverse communities perhaps including the invasive C. raciborskii in clearer waters.

The genetic diversity and the potential toxicity of bloom-forming cyanobacteria were studied in four lagoons located in the state of São Paulo (Campinas, Limeira and Piracicaba cities). Bloom samples were collected on the water surface and cyanobacterial communities were evaluated using DGGE fingerprinting and 16S rDNA clone library. The amplification of genes encoding secondary metabolites such as microcystin (mcy), anatoxin (ana), cylindrospermopsin (cyr), saxitoxin (sxt), cyanopeptolin (mcn) and aeruginosin (aer) was performed and their production analyzed by LC-MS. The comparison of DGGE banding pattern among the different water samples suggested that some operational taxonomic units (OTUs) in these locations were predominant over others. The 16S rDNA clone libraries sequences matched with nine different known cyanobacterial genera available in NCBI, identified as Anabaena, Brasilonema, Cylindrospermopsis, Limnococcus, Microcystis, Nostoc, Pseudanabaena, Synechococcus and Woronichinia. The lagoons ESALQ2, Taquaral and Limeira had more than 80% of the cyanobacterial community assigned to the genus Microcystis. Genes encoding aeruginosin, cyanopeptolin and microcystin synthetases and saxitoxin synthase were amplified, and LC-MS/MS confirmed the production of aeruginosin, cyanopeptolin and microcystin. Rapid and sensitive methods for the detection of these secondary metabolites, especially toxins, using chemical and molecular tools together, can be used for a faster diagnostic of toxic cyanobacterial blooms.
