In this work, a new photoelectrochemical biosensor based on Ag2S nanoparticles (NPs) modified macroporous ZnO inverse opals structure (IOs) was developed for sensitive and rapid detection of alpha fetal protein (AFP). Small size and uniformly dispersed Ag2S NPs were prepared using the Successive Ionic Layer Adsorption And Reaction (SILAR) method, which were adsorbed on ZnO IOs surface and frame work as matrix for immobilization of AFP. The composite structure of ZnO/Ag2S expanded the scope of light absorption to long wavelength, which can make full use of the light energy. Meanwhile, an effective matching of energy levels between the conduction bands of Ag2S and ZnO are beneficial to the photo-generated electrons transfer. The biosensors based on FTO (fluorine-doped tinoxide) ZnO/Ag2S electrode showed enough sensitivity and a wide linear range from 0.05 ng/mL to 200 ng/mL with a low detection limit of 8 pg/mL for the detection of AFP. It also exhibited high reproducibility, specificity and stability. The proposed method was potentially attractive for achieving excellent photoelectrochemical biosensor for detection of other proteins.

GirişAlfa feto protein (AFP) İnsan fetüsünde ilk kez Bergstrand CG ve Czar B tarafından 1956 yılında gösterildi (1). Mide kanserlerinin bazı tiplerinde serum AFP düzeylerinin arttığı ilk kez 1970 yılında bildirilmiştir (2). Bu tip mide kanserleri önceleri adenokarsinom tipi mide kanserlerinin, kötü prognozlu bir varyantı olarak kabul edildi, daha sonraki yıllarda Hepatoid gastrik karsinom olarak ÖZET Amaç: Alfa-fetoprotein (AFP) üreten mide kanserlerinin üretmeyenlere göre daha kötü prognoza sahip olduğu bildirilmektedir. Bu çalışmada, merkezimizdeki AFP üreten mide kanserlerinin sıklığının araştırılması ve üretmeyen tiplerin patoloji sonuçlarının değerlendirilmesi amaçlandı. Gereç ve Yöntem:Mide adenokarsinomu tanısı almış ve ameliyat planı ile genel cerrahi servisine yatırılmış hastalar çalışmaya dâhil edildi. Çalışma 2014-2015 yılları arasında, mide kanseri tanısı almış vakaların verilerinin, prospektif olarak toplanması şeklinde tasarlandı. Hastalardan ameliyat öncesi ve sonrası kan örnekleri alınarak serum AFP düzeyi çalışıldı. Hastane sisteminde kayıtlı patoloji verileri ile AFP düzeylerinin ilişkisi irdelendi. Bulgular:Toplam 63 hasta değerlendirilmeye alındı. AFP yüksekliği 2 hastada saptandı (insidans= %3.17). Serum AFP düzeyi normal sınırlarda olan hastaların, ameliyat öncesi ve sonrası AFP miktarları sırasıyla; 1.520 ng/mL ve 0.590 ng/mL bulundu (p< 0.001). Ameliyat öncesi serum AFP düzeyi ile tümörün evresinin ve nörovasküler invazyon varlığının olup olmaması ilişkisiz bulundu. AFP düzeyi ile metastatik lenf nodu sayısının korelasyonun olmadığı saptandı ( Spearmen's rho= -0.183, p=0.157). Tümörün yerleşim yerine göre AFP düzeyleri arasında anlamalı fark saptandı (p=0.021). Sonuç:Merkezimize başvuran mide kanserleri arasında, AFP üreten tipler çok azdır. AFP düzeyi ile tümörün evresi ilişkisiz gözükmektedir. AFP düzeyi normal sınırlarda olan hastalarda göreceli olarak, distal yerleşimli tümörlerin daha dikkatli değerlendirilmesi önemlidir.Anahtar Kelimeler: Alfa-fetoprotein, mide kanseri, tümör evresi ABSTRACT Objective: It has been reported that Alpha-fetoprotein (AFP) producing gastric cancers have a worse prognosis than those not producing. We aimed to investigate gastric cancers patients in regards of AFP producing cancers incidence and pathological variables of those not producing AFP. Materials and Method:Patients who had diagnosed with gastric adenocarcinoma and hospitalized in general surgery ward for operation were enrolled to the study. Study was designed to collect data of gastric cancer patients in a prospective manner between dates 2014-2015 in a university health center. Patients' blood samples were collected before and after surgery for analyzing AFP. The relation between serum levels of AFP and histopathological variables were analyzed.Results: Totally 63 cases were evaluated. Increased AFP level was observed in 2 cases (incidence=3.17%).
