Alston Virus, a Novel Paramyxovirus Isolated from Bats Causes Upper Respiratory Tract Infection in Experimentally Challenged Ferrets

Abstract: Multiple viruses with zoonotic potential have been isolated from bats globally. Here we describe the isolation and characterization of a novel paramyxovirus, Alston virus (AlsPV), isolated from urine collected from an Australian pteropid bat colony in Alstonville, New South Wales. Characterization of AlsPV by whole-genome sequencing and analyzing antigenic relatedness revealed it is a rubulavirus that is closely related to parainfluenza virus 5 (PIV5). Intranasal exposure of mice to AlsPV resulted in no clinic… Show more

“…The emergence of pathogenic paramyxoviruses, such as Nipah virus (NiV) and Hendra virus (HeV) (1), from native host reservoirs into human populations has driven recent efforts to survey virus prevalence in animals. Such investigations, especially those performed in bat populations (27), have revealed unexpected genomic breadth, expanding the number of genera within the Paramyxoviridae family (8, 9) and highlighting the potential for novel paramyxoviruses to spill over and threaten human health and animal husbandry.…”

“…For next-generation sequencing of whole viral genomes, purified viral RNA was concentrated using a RNA Clean & Concentrator™-5 kit (Zymo Research, Irvine, CA, USA) and RNA concentration was determined using a Qubit RNA HS assay kit read on a DeNovix DS-11FX Spectrophotometer/Fluorometer (DeNovix, Wilmington, DE, USA). Total RNA was reverse-transcribed then isothermally amplified using a REPLI-g WTA Single Cell kit (Qiagen, Hilden, Germany) as previously described (68). Dual barcoded Illumina libraries were prepared with the Nextera XT DNA kit according to the manufacturer’s protocol (Illumina, San Diego, CA, USA).…”

“…The bat-originating Mapuera virus is genetically and antigenically related to the porcine orthorubulavirus La Piedad Michoacán virus, which causes blue-eye pig disease ( 7 9 ). Similarly, Alston orthorubulavirus, which was isolated from bats, is genetically and antigenically closely related to parainfluenza virus 5, which infects humans, monkeys, and dogs ( 10 ). The mumps orthorubulavirus species includes both human and bat mumps viruses, which are considered conspecific due to their high genetic, antigenic, and functional similarities ( 11 ).…”
