Aminolevulinic acid with gold nanoparticles: a novel theranostic agent for atherosclerosis

Abstract: In this study, 5-aminolevulinic acid (ALA) gold nanoparticles (ALA:AuNPs) functionalized with polyethylene glycol (PEG) were synthesized and administered to rabbits to evaluate their use in clinical practice as theranostic agents for atherosclerosis. This was done by measuring the porphyrin fluorescence extracted from the rabbits' blood and feces. An increase in blood and feces porphyrin emission after ALA:AuNP administration suggests that ALA was incorporated by gold nanoparticles, its structure was preserved… Show more

“…The injected ALA/AuNPs accumulated into atheromatous plaques in a rabbit model of atherosclerosis and their ALA was converted to PPIX. The isolated blood and feces of rabbit demonstrated an increase in the fluorescence intensity of porphyrin, indicating stable encapsulation of ALA into gold nanoparticles and further conversion to fluorescent PPIX upon photo-exposure 36 .…”

“…ALA-derived PPIX has been shown to accumulate especially in atherosclerotic plaques, and its fluorescence intensity can be positively correlated with the plaque macrophage content 34 , 35 . To harness the photodynamic properties of ALA-derived PPIX for early diagnosis and therapy of atherosclerosis, Gonçalves et al, formulated PEGylated ALA gold nanoparticles (ALA:AuNPs) with an average size of 18 nm and pH (~5-8.0)-dependent zeta potential (10-40 mV) 36 . The injected ALA/AuNPs accumulated into atheromatous plaques in a rabbit model of atherosclerosis and their ALA was converted to PPIX.…”

“…This behavior causes many cardiovascular diseases [215]. For example, 5-aminolevulinic acid (ALA) AuNPs functionalized with PEG (ALA-AuNPs-PEG) were synthesized and administered to rabbits to evaluate their use in clinical practice as therapeutic agents for atherosclerosis [216]. ALA is used to differentiate between healthy and diseased tissue in the diagnosis of tumors.…”

“…Peng et al [218] showed that PPIX was detected in atheromatous plaques after the intravenous administration of ALA in rabbits. Similarly, Oliveira et al [216] measured the fluorescence of porphyrin extracted from the blood and feces of rabbits after administration of ALA-AuNPs-PEG, detecting an increase in porphyrin emission and the high stability of ALA when attached to the AuNPs-PEG. These results suggested that this method can aid in the diagnosis and treatment of atherosclerosis with good sensitivity.…”

“…Cross‐linked dextran‐coated iron oxide nanoparticle provided 600‐fold efficacy in the destruction of plaque macrophages and substantially reduced skin phototoxicity as compared to conventional photosensitizers. Furthermore, ALA associated with gold nanoparticles has shown increased specificity to atherosclerotic plaques …”
