An efficient temporary immersion system for micropropagation of hybrid hazelnut

Abstract: An efficient protocol for the micropropagation of hybrid hazelnut (Corylus L.) was developed using a temporary immersion system (TIS) for the proliferation of shoots in a liquid medium. Growth and proliferation of shoots as well as the length and number of nodes in developing shoots were significantly improved by combining two forms of chelated iron: ethylenediamine-N,N′-di-(ortho-hydroxyphenyl) acetic acid (FeEDDHA), and FeSO4/EDTA (FeEDTA). The use of TIS with optimized concentrations of FeEDTA and FeEDDHA s… Show more

“…Acclimatization of cherry birch resulted in a 37% survival rate, which is low compared with other woody species such as American elm (90%) (Shukla et al 2012), hazelnut (80%) (Latawa et al 2016), and Betula pendula (95%) (Simola 1985). While the survival rate of micropropagated cherry birch plants was low, the plants that survived were healthy, vigorous, and developed into typical cherry birch plants.…”

“…Among the cytokinins tested, BA was the most effective in cherry birch for shoot elongation and multiplication. Similarly, BA was more effective for shoot proliferation in a variety of other woody plants including American elm (Shukla et al 2012), hazelnut (Latawa et al 2016), and Betula grossa (Ide 1987). In contrast, shoots of Betula pendula had greater elongation in the presence of 2-IP (Jamison and Renfroe 1990), whereas cherry birch failed to proliferate in response to this cytokinin.…”

“…2 was developed to be compatible with both semi-solid culture and liquid based rocker systems [2428], as well as being a suitable size to integrate commercially available RGB LED strips. One of the major limitations of FDM 3D printing with respect to plant tissue culture is that most materials currently used have relatively low melting points and are not suited to heat sterilization or autoclaving.…”

“…By this approach, as outlined before, the liquid medium does not reach the explants forced by air entering the flask but by mechanical movement of the bioreactor (Figure 4). Hazelnut and cedar shoots proliferated more by TIS than by SS, showing higher content of photosynthetic pigments [41], as well as larger leaves and more vigorous shoots [41,42]. Although hazelnut shoots rooted better when adventitious rooting was induced in SS than by TIS without ventilation [41], the rocker system did enhance root formation and acclimation (up to 98%) in the case of Cedrela odorata [42].…”
