An improved simple method for the identification of Mycobacteria by MALDI-TOF MS (Matrix-Assisted Laser Desorption- Ionization mass spectrometry)

Abstract: The aim of this study was to establish a simple method for the rapid identification of Mycobacteria species by MALDI-TOF (Matrix-Assisted Laser Desorption/Ionization-Time of Flight Mass spectrometry) using the Bruker MALDI-TOF Biotyper system (Bruker Daltonik, Bremen, Germany). A multicentre, prospective, and single blind study was performed in three European Hospitals, two Spanish and one UK hospital from May to August 2018. The BD BACTEC MGIT (Becton Dickinson, Berks, UK) liquid culture system was used in al… Show more

“…Such extracts produce different mass spectra that can be used for identification [ 324 ]. This technique was initially developed targeting the identification of members of MTC for the clinical approach but has more recently been expanded to the identification of NTM [ 324 , 325 , 326 ], while databases for spectra analyses are growing.…”

TOF MS technology is applied to the identification of foodborne yeasts and yeast-like fungi[20], filamentous fungi[21], mycobacteria[22], Nocardia[23], and Porphyromonas[24]. Kuhnert et al[25] used MALDI-TOF MS to identify Pasteurellaceae species and subspecies from 250 species in an efficient and simple way.…”

“…Traditionally, the biochemical phenotypic identification of mycobacteria lacks accuracy [ 69 ]. Molecular methods are widely used in clinical laboratories because of their high identification accuracy; however, these methods are time-consuming [ 70 ]. MALDI-TOF MS overcomes the shortcomings of phenotypic and molecular techniques.…”
