Angiopoietin-like protein 3 (ANGPTL3) deficiency and familial combined hypolipidemia

Abstract: Three members of the angiopoietin-like (ANGPTL) protein family-ANGPTL3, ANGPTL4 and ANGPTL8- are important regulators of plasma lipoproteins. They inhibit the enzyme lipoprotein lipase, which plays a key role in the intravascular lipolysis of triglycerides present in some lipoprotein classes. This review focuses on the role of ANGPTL3 as emerged from the study of genetic variants of Angptl3 gene in mice and humans. Both loss of function genetic variants and inactivation of Angptl3 gene in mice are associated w… Show more

“…However we did not find a significantly correlation with TG levels, which is in line with previously reported results 3 , 14 , but not with others showing that circulating ANGPTL3 was negatively correlated with TG levels 38 . The controversial relationship between ANGPTL3 levels and TG suggests that there are other factors affecting the interaction between ANGPTL3 levels and TGs, and further studies are needed to better understand this relationship 3 , 14 , 38 , 39 . In this regard, it is worth pointing out that ANGPTL3 levels are significantly higher in patients with advanced forms of NAFLD or nonalcoholic steatohepatitis (NASH), but no significant differences were detected in individuals with simple fatty liver (NAFL) when compared to healthy and normal weight individuals 20 .…”

“…Circulating cholesterol levels are a function of several concomitant processes such as synthesis, absorption, distribution, uptake and degradation. Circulating factors such as PCSK9 [ 17 20 ] and ANGPTL3 [ 21 , 22 ] also contribute to the regulation of cholesterol levels. The lack of robust association between cholesterolemia and breast cancer might be explained by the interplay between the above-mentioned processes.…”

“…Angiopoietin-like protein 3 (ANGPTL3) is an inhibitor of both lipoprotein lipase and hepatic lipase [ 37 40 ]. Individuals with ANGPTL3 loss-of-function have higher lipoprotein lipase activity and display low blood levels of triglycerides, LDL and HDL-cholesterol, a condition known as familial combined hypocholesterolemia [ 21 , 22 ]. This condition prompted the development of ANGPTL3 inhibitors to treat an almost opposite phenotype: homozygous familial hypercholesterolemia [ 41 43 ].…”

“…In fact, studies on plasma ANGPTL3 concentrations have focused mainly on patients with obesity or cardiovascular disease to elucidate the role of ANGPTL3 in lipid metabolism. 6 , 7 , 23 Research on the role of ANGPTL3 in glucose metabolism has recently been of interest. 13 This is the first study in Vietnam on ANGPTL3 plasma concentrations of diabetic patients.…”
