Angiotensin-converting enzyme-2 overexpression improves atrial electrical remodeling through TRPM7 signaling pathway

Abstract: Atrial electrical remodeling is an important factor in the development and persistence of atrial fibrillation. The aim of this study was to examine the effects of atrial angiotensin-converting enzyme-2 overexpression on atrial electrical remodeling and to elucidate the molecular mechanisms underlying these effects. Twenty-eight male and female dogs were randomly divided into the following 4 groups: a sham-operation group, a control group, an adenovirus-enhanced green fluorescent protein (Ad-EGFP) gene group an… Show more

“…Its presence in ventricular 60 and atrial 61 cardiomyocytes was demonstrated by electrophysiology and mRNA expression. Several studies reported increased TRPM7 basal current levels in human atrial cardiomyocytes dissociated from patients with coronary heart disease or arterial hypertension 62 , speculating about its role in atrial fibrillation, fibrosis 63 and electrical remodelling 64 . Other important TRP channel with preferential ventricular over atrial expression is Trpc3, important in both store-operated and receptor-operated Ca 2+ entry 54 .…”

“…As anti-fibrotic effects, A1-7 has shown the ability to increase the mRNA expression of extracellular signal-regulated kinase-1 (ERK)1/ERK2 (Liu et al, 2010). Moreover, the overexpression of ACE2 has been associated with a reduction in the expression of transient receptor potential melastatin 7, which is a Ca 2+ channel expressed on fibroblasts that can contribute to the fibrogenesis mediated by the transforming growth factor (TGF) (Zhou et al, 2017). In opposition, ACE2 knockout animal models showed a worse left ventricular remodeling in response to the AII-induced acute injury, suggesting a protective role of non-classic RAAS in the myocardium recovery (Kassiri et al, 2009).…”

“…In a canine model of AF, epicardial delivery of the angiotensin-converting enzyme-2 (ACE2) gene was associated with reduced AF inducibility due to a positive effect on electrophysiological properties of atrial fibroblasts [ 147 ].…”
