Alcoholic and aqueous extracts were obtained from red sweet pepper (Capsicum annuum L.) by different methodologies to evaluate their cytotoxic, antioxidant and antimicrobial properties. Alcoholic extracts (MFP, MSd, SFP, SDP, SSd) from fresh red sweet pepper (FP) and dry pulp (DP) and seed (Sd) were obtained by maceration (M) and Soxhlet (S) equipment using methanol as extraction solvent; whereas aqueous extracts (LFP, LSd) were obtained by decoction followed by lyophilization (L). Human tumoral cell lines from breast (MCF-7 and SKBr3), prostate (PC3) and cervix (HeLa), and fibroblasts (as control) were used to determine the cytotoxic properties by the MTT assay. Antioxidant and antimicrobial properties were determined by DPPH and disc diffusion method, respectively. The extracts SDP and SFP showed the higher cytotoxic activity. The SDP extract had a significant (P < 0.05) in-vitro effect on HeLa (1.9 ± 1.4 µg/mL) and PC3 (< 1 µg/mL) cells with a moderated impact on fibroblasts (26.1 ± 1.2 µg/mL); whereas, SFP had a significant (p < 0.05) effect on MCF-7 cell line (2.1 ± 1.2 µg/mL) with a moderated impact on fibroblasts (25.9 ± 1.0 µg/mL). The higher antioxidant activity was found for MFP (80.3 ± 0.2%) and SFP extracts (75.5 ± 0.5%). Mild antimicrobial activity was only observed for alcoholic extracts. The results showed the potential of red sweet pepper (C. annuum L.) as a source of antioxidant and cytotoxic compounds, and suggest the need of further studies to isolate and characterize the bioactive compounds that impart those properties.

This study describes the chemical composition and antimicrobial activity of an essential oil from the leaves of Croton rhamnifolioides Pax&Hoffm., an aromatic plant widely distributed in northeastern Brazil and commonly used in folk medicine. Eight components were identified: 1.8-cineole (46.32%) as the major compound, followed by 1-felandreno (16.70%), p-cymene (10.21%), sabinene (8.14%) and trans-caryophyllene (4.81%). The essential oil exhibited antibacterial activity against Aeromonas hydrophila, Escherichia coli, Listeria monocytogenes, Salmonella Enteritidis and Staphylococcus aureus, with minimum inhibitory concentrations and minimum bactericidal concentrations ranging from 2.5 to 20 and 5 to 40 µL.mL -1 , respectively. In addition, the essential oil also affected the cellular viability of these bacteria. The results suggest the need for research to establish the potential uses of this essential oil as a natural antimicrobial in the food industry. Key words: Croton rhamnifolioides, essential oil, antimicrobial activity, chemical composition ResumoO presente estudo descreve a composição química e a atividade antimicrobiana do óleo essencial das folhas de Croton rhamnifolioides Pax & Hoffm, uma planta aromática amplamente distribuída no Nordeste do Brasil e largamente utilizada na medicina popular. Cromatografia gasosa -espectrometria de massa foi utilizada para determinar sua composição. Oito componentes foram identificados, sendo 1,8-cineol (46,32%) o composto majoritário, seguido por 1-felandreno (16,70%), p-cimeno (10,21%), sabineno (8,14%) e trans-cariofileno (4,81%). O óleo essencial apresentou atividade antibacteriana contra Aeromonas hydrophila, Escherichia coli, Listeria monocytogenes, Salmonella Enteritidis e Staphylococcus aureus com valores de concentração inibitória mínima (CIM) e concentração bactericida mínima (CBM) entre 2,5 -20 μL/mL e 5 -40 μL/mL, respectivamente. Além disso, o óleo essencial também afetou a viabilidade celular dessas bactérias. Os resultados sugerem a necessidade de estudos para estabelecer o uso potencial deste óleo essencial como antimicrobiano natural na indústria de alimentos. RESUMO: O uso popular de plantas medicinais continua sendo de grande interesse para a população, principalmente em países em desenvolvimento, nos quais o acesso à medicina alopática ainda é muito restrito. Assim, além de sua importância na medicina tradicional, o conhecimento das plantas pode fornecer para a indústria farmacêutica, direta ou indiretamente, princípios ativos de interesse terapêutico. No presente trabalho foi avaliada a atividade antimicrobiana dos extratos metanólicos secos de Croton pulegioides Baill. obtidos a partir da raiz, do caule e das folhas nas concentrações de 25, 50 e 100mg/mL utilizando o método de difusão em Agar (técnica de poços) frente a cinco cepas Gram-positivas, três cepas de fungos e três cepas de bactérias Gram-negativas. Na execução do estudo foi utilizado o antibiótico Gentamicina (100µg/mL) no teste com bactérias, e Cetoconazol (300µg/mL) para as Candidas verificando-se, ao mesmo tempo, a ausência de atividade para o diluente DMSO 20% em água. Na observação dos resultados evidenciaram-se halos de inibição indicando atividade dos extratos metanólicos da raiz, caule e folhas, nas concentrações de 50 e 100mg/ml, frente Bacillus subtilis AM 04; dos extratos metanólicos da raiz e do caule, nas concentrações de 50 e 100mg/ml, frente Staphylococcus aureus AM 103 (ATCC 6538); e do extrato metanólico do caule, nas concentrações de 50 e 100mg/ml, frente S. epidermidis AM 235. Por outro lado, todos os extratos foram inativos nas concentrações avaliadas frente às bactérias Gram-negativas utilizadas no estudo (Pseudomonas aeruginosa AM 206, Escherichia coli AM 1050 e Klebsiella pneumoniae AM 410). Referente à atividade sobre fungos, os extratos metanólicos da raiz e do caule inibiram, nas três concentrações avaliadas, o crescimento de Candida tropicalis AM 1181 e C. albicans AM 1140, enquanto o extrato das folhas, inibiu o crescimento de C. tropicalis AM 1181 nas concentrações de 50 e 100mg/ml e de C. albicans AM 1140 apenas na concentração de 100mg/mL. Finalmente o extrato metanólico das folhas, na concentração de 100mg/mL, foi o único a inibir o crescimento de Candida krusei. Os resultados preliminares apresentados pelo presente trabalho sugeriram que a espécie Croton pulegioides Baill. apresentou resultados promissores quanto ao potencial antimicrobiano.
