Antibiotika aus Basidiomyceten, XII. Cristatsäure, ein modifiziertes Farnesylphenol aus Fruchtkörpern von Albatrellus cristatus

Abstract

Fruchtkdrper von Albatrellus cristutus enthalten in hoher Konzentration Cristatsaure (3), einFarnesylphenol, dessen Seitenkette durch Einbau eines Furanringes modifiziert ist. In einer nahe verwandten amerikanischen Albatrellus-Art kommt anstelle von 3 Grifolsaure (6) vor. Die biologische Aktivitat von 3,6 und verwandten Meroterpenoiden wird untersucht, wobei besonders die cytotoxische Wirkung von Dimethylcristatsaure-methylester (4) bemerkenswert ist. A closely related American Albatrellus species contains grifolic acid (6) instead of 3. Biological screening of 3, 6 and related meroterpenoids revealed remarkable cytotoxic activities especially for dimethylcristatic acid methyl ester (4). AusFruchtkorpern von ,, Grifola confluens" isolierten japanische Forscher 1950 ein schwach wirksames Antibiotikum, Grifolin ' I, dessen korrekte Konstitution 1 einige Jahre spater ermittelt wurde3). Kiirzlich wurde gezeigt, dal3 1 und verwandte Farnesylphenole in Pilzen der Gattung Albatrellus ~o r k o m m e n~.~) , so da8 an der Identitat des japanischen Pilzes mit dem Semmelporling, A . confluens, kein Zweifel besteht . In der vorliegenden Mitteilung berichten wir iiber die Inhaltsstoffe des Griinen Kammporlings, A . cristatus (Schaeff. ex Fr.) Kotl. et Pouz. Setzt man die Soxhlet-Extraktion mit Methanol fort (Sh), so fallen nach dem Erkalten farblose Kristalle aus. Nach Absaugen wird aus Methanol umkristallisiert. Ausb. 1.88 g (9%), Schrnp. 166°C. Nach Misch-Schmp., IR-Spektrum und [ u ] , identisch mil D-Mannit. Isolierung tion Afrornenrin: 20 g gefriergetrocknete Fruchtkorper von A . cristafus (Bayern) uerden rnit Methanol extrahiert. Man dampft die Extrakte ein und isoliert nach mehrfacher Chromatographie des Ruckstands an Sephadex LH-20 (Eluent: Methanol) 22 mg (0.1 %) Atromcntin, nach IR-und DC-Vergleich identisch mit der authentischen Verbindung Crisfafin (2): 150 mg 3 werden in 20 rnl wasserfreiem Pyridin 90 min unter Ruckflul3 erhitzt. Man entfernt das Pyridin i. Vak. und dampft in1 Rotationsverdampfer noch dreimal nach Zugabe von etwas Toluol ein. Der Ruckstand wird zwischen IOproz. Citronensaurelosung und Essigester verteilt, die organische Phase mit Wasser gewaschen und getrocknet. Nach Eindarnpfen verbleibt ein oliger Ruckstand, der aus Methylenchlorid/Petrolether (40 -60°C) kristallisiert. Ausb. 47 mg (35.4%). Schrnp. 4 6~4 8°C ; R , = 0.70(Fleck farbt sich uber HCI blaugrun).
