In the field of oncology, the plant kingdom has an inexhaustible supply of bioactive compounds. Phytochemical compounds isolated from Helleborus species have been found to be useful in various chronic diseases. This has brought Helleborus to the attention of medical researchers. H. purpurascens is a plant characteristic of the Carpathian area, known since ancient times for its beneficial effects. The aim of the study was to evaluate the flavonoids composition of a hydroalcoholic extract of H. purpurascens, as well as to assess its antioxidant activity and antitumor potential at the level of two healthy cell lines and four tumor cell lines. In addition, the expression of the genes involved in the apoptotic process (Bcl-2, Bad, and Bax) were evaluated. The results indicated that the extract has a high concentration of flavonoids, such as epicatechin, quercetin, and kaempferol. The extract has an increased antioxidant activity, very similar to that of the standard, ascorbic acid and cytotoxic effects predominantly in the breast cancer cell line, being free of cytotoxic effects in healthy cell lines. Underlying the cytotoxic effect is the induction of the process of apoptosis, which in the present study was highlighted by decreasing the expression of anti-apoptotic genes (Bcl-2) and increasing the expression of pro-apoptotic genes (Bad and Bax). In conclusion, the hydroalcoholic extract of H. purpurascens can be considered an important source for future medical applications in cancer therapy.

Povećanje incidencije otrovanja biljnim i životinjskim toksinima tijekom proteklih godina ukazuje na važnost poznavanja otrovnih vrsta na području Republike Hrvatske. Od otrovnih biljnih vrsta u Republici Hrvatskoj raste velebilje, kranjski bijeli bun, duhan, bunovina, crna bunika, bijeli kužnjak, crni kukurijek i jesenski mrazovac. Većina otrovnih biljaka pripada porodici pomoćnica (Solanaceae). To su biljke bogate tropanskim alkaloidima koji djeluju kao antikolinergične tvari. Imaju antagonističko djelovanje na muskarinske acetilkolinske receptore. S obzirom na široku rasprostranjenost navedenih receptora u ljudskom organizmu, pri intoksikaciji tim biljkama dolazi do poremećaja rada srca, inhibicije ili stimulacije lučenja pojedinih žlijezda, kao i poremećaja intestinalnog motiliteta. Jedan je od najpoznatijih učinaka pojava delirija i halucinacija koji nastaju zbog inhibicije muskarinskih acetilkolinskih receptora u mozgu. Opisane su i otrovne gljive, iako one pripadaju zasebnome carstvu. U našem području najvažnije su zelena pupavka i muhara. Ove gljive pripadaju rodu Amanita u kojima nalazimo dvije grupe toksina, falotoksine i amatoksine koji inhibiraju RNA polimerazu II i time onemogućuju sintezu vitalno važnih bjelančevina. Od životinjskih toksina opisani su toksini zmija, paukova i riba. Većina zmija otrovnica u Republici Hrvatskoj pripada porodici Viperidae. Naše su najpoznatije otrovnice poskok i riđovka. Njihov otrov sadrži hijaluronidaze, fosfolipazu A2 i metaloproteinaze. Od paukova opisani su crna udovica i smeđi primorski riječni pauk. Otrov pauka smjesa je monoamina, koji djeluju proalgetski, peptida s ekscitatornim učincima koji izazivaju nociceptivni odgovor. Završni dio osvrće se na ribe otrovnice, škrpinu i morskog pauka.

Tinospora sagittata (Oliv.) Gagnep. is an important medicinal tetraploid plant in the Menispermaceae family. Its tuber, namely "Radix Tinosporae" used in Traditional Chinese Medicine, is rich in medicinal terpenoids and benzylisoquinoline alkaloids (BIAs), To enhance understanding the biosynthesis of medicinal compounds, we, herein, report the assembly of a high quality chromosome-scale genome with both PacBio HiFi and Illumina sequencing technologies. The size of assembled genome was 2.33 Gb consisting of 4070 scaffolds (N50=42.06Mb), of which 92.05% were assigned to 26 pseudochromosomes in A and B sub-genomes. A phylogenetic analysis with the T. sagittata and other 16 plant genomes estimated the evolutionary placement of T. sagittata and its divergence time in Ranunculales. Further genome evolution analysis characterized one round tandem duplication about 1.5 million years ago (MYA) and one whole-genome duplication (WGD) about 86.9 MYA. WGD contributed to the duplication of clade-specific cytochrome P450 gene family in Ranunculales. Moreover, sequencing mining obtained genome-wide genes involved in the biosynthesis of alkaloids and terpenoids. TsA02G014550, one candidate, was functionally characterized to catalyze the formation of (S)-canadine in the jatrorrhizine biosynthetic pathway. Taken together, the assembled genome of T. sagittata provides useful sequences to understand the biosynthesis of jatrorrhizine and other BIAs in plants.
