Antifungal activity of 4-hydroxy-3-(3-methyl-2-butenyl)acetophenone against Candida albicans: evidence for the antifungal mode of action

Abstract: The main secondary metabolite of Senecio nutans is 4-hydroxy-3-(3-methyl-2-butenyl)acetophenone (4HMBA). The antifungal activity of this compound and three derivatives was assessed using Candida albicans. 4HMBA exhibited the highest antifungal activity among the assayed compounds. The Fractional Inhibitory Concentration (FIC = 0.133) indicated a synergistic fungicidal effect of 4HMBA (5 mg L(-1)) and fluconazole (FLU) (0.5 mg L(-1)) against the C. albicans reference strain (ATCC 10231). Microscopy showed that … Show more

“…The closely-related compounds furfuryl alcohol and acetyl furan, as well as three of the six phenyl-ketones tested also performed well against P. infestans germination. Acetophenone derivatives are well-described antifungals thought to target the fungal cell wall (Soberon et al, 2015 ; De Aguiar et al, 2016 ). A high level of inhibition also resulted from exposure to diphenylamine and 2,5-dimethylpyrazine treatments; however we cautiously consider these compounds as artifacts originating from the medium.…”

“…Apesar de ainda não haver dados sobre a atividade antifúngica do EPHO, outras moléculas da classe das acetofenonas e derivados já demonstraram atividade contra diferentes fungos, incluindo C. albicans, sugerindo também que sua ação farmacológica provavelmente ocorra devido a inibição da formação da parede celular fúngica além de impedir o desenvolvimento de estruturas de virulência como pseudohifas [19,32].…”

“…For example, Candida albicans and Aspergillus fumigatus are opportunistic fungi. These fungi can produce invasive fungal infections in any organ of humans [3]. Limited numbers of therapeutic antifungal agents are available for these fungi because of increased resistance mechanisms through the biofilms' formation by the fungal strains [2].…”
