Antioxidant, Hepatoprotective Potential and Chemical Profiling of Propolis Ethanolic Extract from Kashmir Himalaya Region Using UHPLC-DAD-QToF-MS

Abstract: The aim of this study was to examine hepatoprotective effect of ethanolic extract of propolis (KPEt) from Kashmir Himalaya against isoniazid and rifampicin (INH-RIF) induced liver damage in rats. Hepatic cellular injury was initiated by administration of INH-RIF combination (100 mg/kg) intraperitoneal (i.p.) injection for 14 days. We report the protective effects of KPEt against INH-RIF induced liver oxidative stress, inflammation, and enzymatic and nonenzymatic antioxidants. Oral administration of KPEt at bot… Show more

“…CSP and its flavonoids demonstrated significant prophylactic impact as a result of antioxidant activity and is apparently seen in histological findings of liver sections with distinct hepatocytes, sinusoidal spaces, central veins, and mild degrees of fatty change, necrosis infiltration almost comparable to the group III group. These findings are in accordance with the earlier reports stating the role of flavonoids against hepatotoxicity and oxidative stress [25,54]. Previous studies have also established that flavonoids reverse INH-RIF-induced fibrosis and necrosis [55].…”

“…The DPPH and ABTS assay are the most common method used to assess the radical scavenging ability of the different compounds because they have the capacity to give hydrogen free radicals. The mechanism behind the radical scavenging capacity is due to flavonoids and phenolic acids, and these are generally weak in nature, and thus act as competent electrons capable of responding to O2 ш-depending on the substitution within the phenolic ring [25].…”

“…UHPLC-MS has been used widely for analyzing propolis (Bankova et al 2000, Sawaya et al 2011, Novak et al 2014, Wali et al 2015, Ristivojević et al 2015, Sagi et al 2016, but it has limitations in terms of the identification of uncommon substances because there is no database for making comparisons among geopropolis samples. Therefore, the unambiguous characterization of the compounds corresponding to the ions at m/z 601, 475, and 569 may be possible after their isolation and further analysis using other spectrometric methods (e.g., NMR), in addition to MS experiments.…”

“…A significant increase in serum activities of enzymes alanine aminotransferase (ALT), and aspartate aminotransferase (AST) was observed in MTX treated group which are markers of liver inflammation and are the major critical liver enzymes involved in the metabolism of amino acids and electron transport chain [27,28]. Treatment with the dual extracts modulated the effect of MTX, bringing about a recovery in the elevated enzyme activities comparable to the effect of silymarin.…”
