Applying liquid chromatography-tandem mass spectrometry to assess endodontic sealer microleakage

Abstract: The objective of this study was to describe a new method for the quantitative analysis of a microleakage of endodontic filling materials. Forty extracted single-rooted teeth were randomly divided into three experimental groups. After root canal shaping, the experimental groups were filled using the lateral condensation technique with the Epiphany system (G1), with gutta-percha + Sealapex (G2), and with gutta-percha + AH Plus (G3). Each root was mounted on a modified leakage testing device, and caffeine solutio… Show more

“…Caffeine microleakage is another method with the use of a modified dual-chamber model; epoxy resin is used for sealing of the margins instead of cyanoacrylate glue and leaked caffeine content can be detected by high-performance liquid chromatograp-hy-tandem mass spectrometry (HPLC-MS/MS) (83) or a visible ultraviolet spectrophotometer (UV-VIS detectors) (77,84). The caffeine detection limit was 2000 ng mL-1, using UV-VIS, and 10 ng mL-1, using HPLC-MS/MS.…”

“…Even in the Iranian population, 95.4% BS in the RNs and CC in mandibular canines was noted [ 22 ]. In a Turkish subset, the BS of RNs and CNs for mandibular anterior teeth were 96–100% and 90–95% [ 32 ]. In this analysis, mandibular canines showed a high BS for the RNs (97.8%), CNs (74.8%), and CCs (67.77%).…”
