Immunosensors are solid-state devices in which the immunochemical reaction is coupled to a transducer. They form one of the most important classes of affinity biosensors based on the specific recognition of antigens by antibodies to form a stable complex, in a similar way to immunoassay. Depending on the type of transducer there are four types of immunosensor: electrochemical, optical, microgravimetric and thermometric. The most commonly used bioelements for the development of electrochemical immunosensors are antibodies (Ab), followed by aptamers (Apt) and, in the last five years, microRNA (miRNA). In order to perform an early diagnosis, a method that is able to measure peptides and proteins directly in a sample, without any sample pre-treatment or any separation, is preferred. This direct detection can be performed with methods making use of the specific interaction of proteins with Ab, Apt and miRNA. The recent developments made in the immunosensor field, regarding the incorporation of nanomaterials for increased sensitivity, multiplexing or microfluidic-based devices, may have potential for promising use in industry and clinical analysis. Some examples of assays for several commercially available biomarkers will be presented. The main application fields, beside biomedical analysis, are drug abuse control, food analysis and environmental analysis.

Pharmacokinetics of recombinant human basic fibroblast growth factor (rhbFGF) in mice were studied by using a radioiodination method combined with sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and a sandwich enzyme-Linked immunosorbent assay (ELISA). RhbFGF concentration in serum were dertermined after iv administration of 2.5, 5 and 10 microg/kg of 125I-rhbFGF or rhbFGF. The AUCs were linearly correlated to doses. Concentrations in tissues, recoveries of 125I-rhbFGF in urine and feces were also analyzed following iv dose of 5 microg/kg. Pharmacokinetic profiles of radiolabelled and unlabelled rhbFGF were consistent in trend but exit certain differences. Mice cleared unlabeled rhbFGF markedly faster than their radiolabeled counterparts.

Recebido em 4/1/02; aceito em 27/6/02 SPR -NEW TOOL FOR BIOSENSORS. This paper reviewed the development and theoretical aspects of surface plasmon ressonance (SPR) technique and discusses this powerful sensor technology in the development of biosensors, as well as for the investigation of biological interactions and clinical assays. The SPR has been proven to be a valuable tool to investigate dynamic processes, such as adsorption, degradation, determination of dieletric properties, association/dissociation kinetics, affinity constants of specific ligand-ligate interactions, allowing real-time analysis at almost any surface. The SPR as a complementary technique alongside electrochemical methods is also presented.
