Autocrine growth factors in human periodontal ligament cells cultured on enamel matrix derivative

Abstract: Cultured PDL cells exposed to EMD increase attachment rate, growth rate and metabolism, and subsequently release several growth factors into the medium. The cellular interaction with EMD generates an intracellular cAMP signal, after which cells secrete TGF-beta1, IL-6 and PDGF AB. Epithelial cell growth however, is inhibited by the same signal. This suggest that EMD favours mesenchymal cell growth over epithelium, and that autocrine growth factors released by PDL cells exposed to EMD contribute to periodontal … Show more

“…Yet, little is known about the critical factors promoting MSCs expansion in the co-culture system. Tendon/ligament cells were reported to secrete a number of growth factors when healing or stimulated, such as IGF, TGF-b, FGF-2 and PDGF (Lyngstadaas et al 2001;Dahlgren et al 2005), among which FGF-2, PDGF and BMP-2, a member of TGF-b superfamily, have been illustrated to promote MSCs division in vitro (Martin et al 1997;Liu et al 2009;Choi et al 2008;Tamama et al 2006). In our co-culture system, the exact factors playing the critical role in MSCs proliferation need to be further clarified.…”

“…This finding corroborates with other EMD related studies. 23 EMD medium led to a higher BSP gene expression irrespective of stretching. Collagen I expression was lower in the stretched group, but this was compensated by the effect of EMD.…”

“…TGF-β 1 is known to be related to the mitogenesis of cells and the remodeling of extracellular matrix 12,14) while FGF-2 has a potent angiogenic effect 15) . IL-6 is one of the well-known and plentiful inflammatory cytokines 16) . In this regard, Koh et al 17) studied the cytokine production from osteoblastic cells in contact with MTA, and they found that MTA has an up-regulating effect on the expression of IL-1α , IL-1β , and IL-6 from osteoblasts.…”
