Beneficial effects of tempol, a membrane-permeable radical scavenger, in a rodent model of collagen-induced arthritis

Abstract: This study is the first to provide evidence that tempol, a small molecule that permeates biologic membranes and scavenges reactive oxygen species, attenuates the degree of chronic inflammation and tissue damage associated with CIA in the rat.

“…Each paw was scored macroscopically, based on redness/ swelling/deformity of the joint (35): 0 ϭ no signs of arthritis, 1 ϭ swelling and/or redness of the paw or 1 digit, 2 ϭ 2 joints involved, 3 ϭ Ͼ2 joints involved, and 4 ϭ severe arthritis of the entire paw and digits (36). An arthritis index score for each rat was calculated by adding the scores for the individual paws.…”

“…Tyrosine nitration, an index of the nitrosylation of proteins by ONOO Ϫ and/or oxygen-derived free radicals, was determined by immunohistochemistry as previously described (36). On day 35, the joints were trimmed, placed in decalcifying solution for 24 hours, and 8-m sections were prepared from paraffin-embedded tissues.…”

“…It has been hypothesized that nitroxides could decrease the inflammatory response and attenuate the damage caused by ROS released by immune effector cells. Tempol has been preliminarily shown to be effective in several models of inflammation including pancreatitis [49], pleurisy [76], arthritis [77], colitis [78], and uveoretinitis [79] and more studies are ongoing.…”

“…Specific supplementation of oral vitamin E, the major lipid-soluble antioxidant in human plasma, erythrocytes, and tissue, had no effect on RA disease activity or indices of inflammation but did improve pain, suggesting a role in central analgesia mechanisms (Edmonds et al, 1997). Various forms of antioxidant therapy have demonstrated promising results in experimental arthritis models (Bandt et al, 2002;Cuzzocrea et al, 2000;Venkatraman & Chu, 1999).…”
