Bidentate Zinc Chelators for α‐Carbonic Anhydrases that Produce a Trigonal Bipyramidal Coordination Geometry

Abstract: A series of new zinc binding groups (ZBGs) has been evaluated kinetically on 13 carbonic anhydrase (CA) isoforms. The fragments show affinity for all isoforms with IC50 values in the range of 2–11 µm. The crystal structure of hCA II in complex with one such fragment reveals a bidentate binding mode with a trigonal-bipyramidal coordination geometry at the Zn2+ center. The fragment also interacts with Thr199 and Thr200 through hydrogen bonding and participates in a water network. Further development of this ZBG … Show more

“…The crystal structure of hCAII L198G with 1,2-HOPTO bound (1.42 Å resolution) shows the MBP does not change its binding geometry when compared to the same compound bound in wild type hCAII (Figure 5, PDB:3M1K) [37]. In both enzymes, 1,2-HOPTO is bound in a bidentate fashion to the Zn(II) ion, with Zn-S and Zn-O bond distances of ~2.4 Å to ~2.6 Å creating a 5-coordinate species at the metal center with a trigonal bipyramidal geometry.…”

“…53 Finally, resorcinol ( 3 ) has been shown to inhibit hCAII by hydrogen bonding to the Zn(II)-bound hydroxide, and does not engage in metal ion binding. 54 Inhibition constants and previously reported binding modes for these molecules are summarized in Table 2.…”

“…[1][2][3][4] All the zinc binders known so far (organic compounds) coordinate monodentately to the zinc ion through a nitrogen atom from the zinc-binding group (ZBG), 7-10 whereas few bidentate inhibitors have been discovered, such as a thiomalthol anlogue (HOPTO) 11 and N-hydroxyurea 12 which were shown to coordinate bidentately to the Zn(II) ion from the enzyme active site. Dithiocarbamates (DTCs) are well known metal complexing agents and they also possess interesting biomedical and agricultural applications.…”
